Morphological assessment of the behavior of three‐dimensional anionic collagen matrices in bone regeneration in rats

Abstract: The osteogenic behavior of three-dimensional collagenic and anionic matrices (M3DGA) implanted in critical bone defects of 8 mm diameter was morphologically assessed. The defects were performed in the calvaria of 48 adult male (Wistar) rats, and observed at days 15, 30, and 60. The animals were distributed in four groups of equal number: GA1 (M3DGA with 60 min of cross-linking in glutaraldehyde [GA]); GA2 (M3DGA with 30 min of cross-linking in GA); GA3 (M3DGA with 15 min of cross-linking in GA); and G4 (contro… Show more

“…Critical defects do not regenerate during the life span of the animal [12,26,[34][35][36]; thus they are a good model for the evaluation of the osteogenic potential of biomaterials designed to be used in bone regeneration. In our work we used 8 mm for the diameter of the defect in the calvaria of rat.…”

“…Each group of animals was divided into two subgroups for euthanasia at either 15 or 60 days postoperative, in a total amount of five animals/biomaterial/period of observation. These periods of observation were chosen since they represent the initial inflammatory response to the biomaterial implantation (15 days) and the consolidated aspect of the implanted region (60 days) 28. The area of the original surgical defect and the surrounding tissues were removed en bloc .…”

The blocks were fixed in 10% neutral formalin, rinsed with water and then decalcified in Morse's acid solution.…”

“…Bone neoformation generally depends on the size and type of the defect [Takagi and Urist, 1982;Schmitz and Höllinger, 1986;Chesmel et al, 1998, Miguel et al, 2006, the species of the animal and the implant site. However, repairing defects of critical dimensions is still very difficult, because under these conditions, there is not sufficient formation or organization of blood clots [Frame, 1980], which is essential for bone regeneration.…”
