Prorocentrum mexicanum and P. rhathymum are toxicologically important dinoflagellates, but their relationship is not well defined. We investigated strains from Puerto Rico and Brazil by light and scanning electron microscopies. We provide molecular data from a strain isolated near the type locality of P. rhathymum, and the first morphological and molecular data from the South Atlantic Ocean. The rRNA gene (rDNA) sequences of the Puerto Rican and Brazilian strains were identical, and their morphologies fit the description of P. rhathymum. In the molecular phylogenies, the globally distributed populations under the names P. mexicanum and P. rhathymum are intermixed and branched together, except for several divergent strains from Florida and Cuba. We examined the original descriptions and iconotypes of the species Prorocentrum maximum, P. brochii, P. mexicanum, and P. rhathymum. We conclude that P. maximum sensu Schiller's figure 41a corresponds to the earlier description of this species; the split of P. mexicanum and P. rhathymum was based on a misidentification because P. mexicanum sensu Cortés-Altamirano & Sierra-Beltrán corresponds to P. texanum var. cuspidatum; and P. rhathymum is a junior synonym of P. mexicanum. Several Floridian and Cuban strains correspond to a new species, which we describe as Prorocentrum steidingerae sp. nov.

The resting cysts of the benthic dinoflagellate Prorocentrum leve from a natural reservoir in Gujan-Mestras (Gironde, France) were described in this study. The incubated urn-shaped cysts gave rise to cells of P. leve. Morphological observations through light microscopy and scanning electron microscopy, particularly of the periflagellar platelets, combined with large subunit ribosomal DNA sequences obtained through single-cell analysis confirm their affinity to the species P. leve. The cysts were characterized by a specific shape and the presence of an anterior plug. This is the first conclusive evidence for fossilizable resting stages within the Prorocentrales, one of the major orders within the Dinophyceae. Palynological treatments show that the cysts and endospores withstand hydrochloric and hydrofluoric acids. Micro-Fourier transform infrared analysis on single specimens suggests that the composition of the endospore is cellulosic and the cyst wall a more robust, noncellulosic β-glucan. The spectra overall are similar to other published spectra of resting cysts from autotrophic, planktonic dinoflagellates.

A B S T R A C TThirteen isolates of Prorocentrum species were established from the coral reefs of Perhentian Islands Marine Park, Malaysia and underwent morphological observations and molecular characterization. Six species were found: P. caipirignum, P. concavum, P. cf. emarginatum, P. lima, P. mexicanum and a new morphotype, herein designated as P. malayense sp. nov. Prorocentrum malayense, a species closely related to P. leve, P. cf. foraminosum, P. sp. aff. foraminossum, and P. concavum (Clade A sensu Chomérat et al. 2018), is distinguished from its congeners as having larger thecal pore size and a more deeply excavated V-shaped periflagellar area. Platelet arrangement in the periflagellar area of P. malayense is unique, with the presence of platelet 1a and 1b, platelet 2 being the most anterior platelet, and a broad calabash-shaped platelet 3. The species exhibits consistent genetic sequence divergences for the nuclear-encoded large subunit ribosomal RNA gene (LSU rDNA) and the second internal transcribed spacer (ITS2). The phylogenetic inferences further confirmed that it represents an independent lineage, closely related to species in Clade A sensu Chomérat et al. Pairwise comparison of ITS2 transcripts with its closest relatives revealed the presence of compensatory base changes (CBCs). Toxicity analysis showed detectable levels of okadaic acid in P. lima (1.0-1.6 pg cell ˗1 ) and P. caipirignum (3.1 pg cell ˗1 ); this is the first report of toxigenic P. caipirignum in the Southeast Asian region. Other Prorocentrum species tested, including the new species, however, were below the detection limit.
