Morphological and growth altering effects of Cisplatin in C. albicans using fluorescence microscopy

Abstract: Changes in morphology and growth curve of Candida albicans in response to treatment by Cisplatin has been studied using fluorescence staining with ethidium bromide. Treatment with Cisplatin was found to markedly inhibit hyphae and ovoid growth as revealed by ethidium bromide staining of drug treated cells. These changes were concomitant with inhibitory effects on the growth curve with respect to untreated cellsPresence of Cisplatin not only caused suppression in the limiting values in the growth curve, but als… Show more

“…C. albicans SC5314 was cultured in RPMI 1640 medium containing 11g (0, 0.5, and 1 × 10 –3 mg/mL) with an inoculums size of 10 6 cells/mL. Cells were cultured at 30°C with constant shaking and optical density at 600 nm were measured after incubating for 0, 1, 2, 4, 8, 12, 16, 24, and 32 h ( Kesavan et al, 2005 ). Three independent experiments were performed.…”

“…Identifying the mid-log phase is important to identify as it is the time point to compare a kānuka treatment condition to an untreated condition. Figure 3.10 shows the normal growth of P. acnes wherein exponential growth occurs for 60 hours with mid-log at 32 hours, which is consistent with other reputable studies (Kesavan et al, 2005;Lopez-Abarrategui et al, 2016).…”
