Research was conducted at Al-Qasim Green University in the Plant Tissue Culture Laboratory in the Department of Horticulture and Landscaping in the College of Agriculture from November 2021 to May 2022 with the aim of comparing two types of cytokinins: BAP at a concentration of 2.0 mg/L-1 and TDZ at a concentration of 0.2 mg/L-1 and two concentrations Iron (2.5-5 mg/L-1). To know their effect on the average of shoot multiplication in vitro of the root and its content of protein and macro (N.K) and Micro (iron) elements. The research was implemented as a factorial experiment using a complete random design (CRD) with two factors: cytokinin and nano iron. The results indicated a superiority of 0.2 mg.L-1 TDZ in each of the Multiplication shoots (4.667 shoots/explant ) and the content of shoots of iron (54.66 mg.kg-1), while BAP at a concentration of 2.0 mg/L-1 was excelled on the content of shoots of nitrogen, potassium and protein (1.778%, 1.947%.) respectively. As for nano iron at a concentration of 2.5 mg. L-1 was significantly excelled in the average of multiplication shoots (5 shoots/explant ), nitrogen (1.765%) and protein (10.98%). Specifically, the 5 mg dose. Potassium and iron (1.692% and 55.0 mg.kg-1) were most abundant in L-1.

Bananas and plantains are among the most important food crops in Central and West Africa. Their plantation is lead to many problems. In the recent decades, biotechnology tools using in vitro culture technics are used for the mass and free disease plantlets production in order to increase the bananas production and the yield. The main way of in vitro tissue culture at this end is the direct organogenesis i.e., the ability of plant tissues to form various organs de novo by shoots or roots induction to differentiate from a cell or cell clusters. This review aims to summarize the main results obtained in the organogenesis of bananas and plantains (Musa spp.) under in vitro conditions and to identify the challenges during the process. The research articles used in this review show that micropropagation is a reliable alternative to conventional production system of bananas and plantains planting material. However, the use of the in vitro micropropagation for bananas and plantains entails choosing the optimal explant type and size according to objectives. Benzylaminopurine remains the preferred cytokinin for in vitro banana and plantain shoot proliferation, while the use of thidiazuron appears to be more and more common. Whichever cytokinin used, the optimal cytokinin concentration for shoot proliferation is genotype dependent. This review also focuses on the causes and control measures of the two major banana and plantain micropropagation constraints: lethal tissues browning/darkening and microbial contaminations. It showed that applying the suitable and available control measure, according to the evolution of culture, is necessary. All this available information on the in vitro conditions makes banana and plantain cultivars in vitro organogenesis possible.

Banana (Musa spp.) is one of the most important fruit crops in the world in terms of consumption and production. Commonly, bananas are propagated using conventional and in vitro techniques. Conventional methods generally involve vegetative propagation by the use of suckers, which is a time-consuming technique and is prone to various diseases. To overcome these problems, tissue culture technique has played a major role. With this technique, plant growth regulators are used to help improve rapid propagation for large-scale production. The plant growth regulators, benzyl adenine (BA) and thidiazuron (TDZ), which are both cytokinins, have gained attention for their effects on plant growth and development in tissue culture. In the present study, the effects of BA and TDZ on the shoot multiplication of Cavendish bananas in vitro were investigated. There were two experiments performed due to their being two types of explants obtained: one-month-old banana plantlets and one-month-old banana clumps. Culture I used one-month-old plantlets, and Culture II used one-month-old clumps. The results revealed that there was no significant difference in shoot formation between BA and TDZ at any concentration. However, the optimum concentration of BA was found to be 3 ppm for plantlets (Culture I), which resulted in 1.60 new shoots, and 4 ppm for clumps (Culture II) producing 4.00 shoots. In the case of TDZ, 2 ppm which produced 2.60 shoots, was the optimum concentration for both types of explants. In addition, the study suggested that clumps were the most suitable choice for banana multiplication. Further investigation focusing on the effects of explant morphology and cutting technique on plant growth is recommended.
