Montelukast, an Anti-asthmatic Drug, Inhibits Zika Virus Infection by Disrupting Viral Integrity

Abstract: The association of Zika virus (ZIKV) infection and severe complications including neurological sequelae especially fetal microcephaly has aroused global attentions since its outbreak in 2015. Currently, there are no vaccines or therapeutic drugs clinically approved for treatments of ZIKV infection, however. And the drugs used for treating ZIKV in pregnant women require a higher safety profile. Here, we identified an anti-asthmatic drug, montelukast, which is of safety profile for pregnant women and exhibited a… Show more

“…A widely used drug among patients with asthma is montelukast, which have bronchodilator and anti-inflammatory effects, it even decreases the need for high doses of steroids ( 23 ). Moreover, montelukast has exhibited antiviral efficacy against ZIKA infection in vitro and in vivo, showing that montelukast could disrupt the integrity of the virions to release the viral genomic RNA, hence irreversibly inhibiting viral infectivity ( 24 ).…”

“…TNF-α, IL-6 and IL-1β levels (66,67). A previous study suggests that it inhibits Zika virus by disrupting the integrity of the virions (68). It has been predicted by other groups as well to bind to main protease of SARS-CoV2 (69).…”

“…For Influenzae A virus, an inhibition of the expression of the viral genome was observed with MK (Landeras-Bueno et al, 2016). For flaviviridae, in particular Zika virus, an irreversible and early inactivation of the virus was reported, probably due to some damage to the lipid membrane (Chen et al, 2020). Three distinct mechanisms were proposed to support the beneficial impact of MK on Zika virus: i) a direct antiviral action, ii) an antagonization of the cytokine storm, and iii) an inhibition of the vertical transmission by a MK-related neuroprotective effect on the brain of fetus.…”
