Monomer‐scale design of functional protein polymers using consensus repeat sequences

Abstract: Protein-based polymers possess chemically defined sequences that can encode diverse properties and functions into a new class of biopolymeric materials. However, sequence variation that emerges from evolution can obscure the sequence-function relationships of naturally derived polymers. One strategy to clarify these relationships is to identify common sequences between proteins with similar functions. These conserved sequences often emerge from repeat proteins, and "consensus repeat sequences" provide a conveni… Show more

“…Multi-site ncAA incorporation is particularly desirable for the design and production of protein-based polymers (PBPs), which consist of tandem repeats of either natural or artificial short peptide motifs, and whose properties are determined by the sequence of the AAs and ncAAs in these motifs ( Connor and Tirrell, 2007 ; Israeli et al, 2019 ; Varanko et al, 2020 ; Chang et al, 2021 ). Although the incorporation of ncAAs in numerous PBPs can be used to expand their range of properties and functions ( Connor and Tirrell, 2007 ; Israeli et al, 2019 ), we focus here on the effect of ncAA incorporation on the properties of two families of bio-inspired, thermo-responsive PBPs: elastin-like polypeptides (ELPs) and resilin-like polypeptides (RLPs).…”

“…Since the establishment of the concept of macromolecule, 1,2 synthesis of new polymers and the study on their structureproperty relationship have always been the focus of polymer science. [3][4][5][6][7][8][9] Controlled synthesis of tailor-made polymers is of great significance in both academia and industry. 10,11 Polyethylene (PE) is arguably the most simple and useful polymer that produced in large amounts.…”
