Monoclonal antibody 9.3 and anti‐CD11 antibodies define reciprocal subsets of lymphocytes

Abstract: We have previously described antibody 9.3 which recognizes a 44-kDa polypeptide designated Tp44 expressed on 70-80% of peripheral blood T cells, including nearly all CD4+ cells and some CD8+ cells. Whereas the 9.3+ population contains helper cells, cytotoxic T cell precursors and cytotoxic T cell effectors, the 9.3- population has been reported in several models to contain precursors for suppressor cells. In this report, we demonstrate that 9.3- lymphocytes express CD11, an antigen which is also present on mon… Show more

“…The CD28 ÿ T cell subset is mostly correspondent to the CD11b or CD57 T cell subset in normal PBMC [14,15]. Increased CD57 or CD11b T cell subset in PBMC has been reported after BMT [21] or in the course of infection by cytomegalovirus [22] and Epstein-Barr virus (EBV) [23].…”

“…In healthy adult peripheral blood, CD28 ÿ T cells were preferentially found within the CD8 subset [14,15], while CD28 ÿ 4 T cells were observed in certain individuals [16]. CD28 ÿ 8 T cells were not observed in the thymus and were present only at low frequency in cord blood.…”

“…B7, a member of the Ig supergene family, has been shown to be a ligand for another member of this family, the T cell surface protein CD28, (10)(11)(12)(13) . CD28 is constitutively expressed on 95% of human CD4+ T cells, 50% of CD8+ T cells, and on thymocytes which coexpress CD4 and CD8 (14)(15)(16) . Following suboptimal activation of T cells with anti-CD3 mAb (16), anti-CD2 mAb, or phorbol ester (17), crosslinking of CD28 by anti-CD28 mAb results in enhanced T cell proliferation and greatly augments synthesis of multiple lymphokines (18).…”

“…Earlier studies on CD28 have indicated that this Ag is variably expressed on most (but not all) T cells from normal adult blood donors, 24,25 whereas essentially all T cells from newborns express CD28, 10,26 but the nature of this discrepancy was unclear. The results from our study partially clarify the nature of the variability of CD28 expression.…”

“…2). 10,24 At the functional level, T cells from normal donors expressing CD57 34 or CD8 and CD11b, 10,35 as well as PBMC of HIV-infected subjects depleted from CD28 + cells, 15 are able to kill targets in an anti-CD3 redirected manner. Our data confirm those previous reports by comparing the cytotoxicity of purified CD3 + CD8 + CD28 7 with CD3 + CD8 + CD28 + and with unseparated CD3 + cells (Fig.…”
