Monoclonal antibodies against two human lung carcinoma cell lines

Abstract: Summary.-Monoclonal antibodies against 2 human lung carcinoma cell lines (E14 and BEN) were prepared by production and cloning of somatic cell hybrids between the murine myeloma NS1, and spleens from E14-and BEN-immune BALB/c mice. Approximately 2000 hybrid culture supernatants were screened for antibody simultaneously against the immunizing cell line and lung fibroblasts (573 Lu) using a radiolabelled Protein A binding assay. Although the vast majority secreted antibodies which recognized species-specific ant… Show more

“…The lysates was then centrifuged at 18000 g for 20 min at 4° C. The protein content of IEE, IEG or IEK cells was quantified by the method of Lowry et al (1951). The polyclonal antibodies against the cell lysates of IEE, IEG or IEK were raised in Swiss white mice using standard immunization protocol (Brown & Moore, 1982). The polyclonal antiserum against lysates of eye, gill or kidney tissue of E. suratensis was raised using the same protocol followed for the production of antibodies against cell lysates.…”

“…We found tissue cultures to be helpful in identifying tumour-related Spots. Additionally tissue cultures are an unlimited source of material for further investigations and have already been used for the production of tumour-associated markers and antibodies (22)(23)(24)(25)(26)(27). Spot A seems to be connected with epithelial tissue and its transforrnation to a carcinoma.…”

“…Several MAbs to non-small-cell lung carcinoma, including squamous-cell carcinoma, large-cell carcinoma, and adenocarcinoma have been described. Most of these antibodies have a broad range of reactivity among lung carcinomas or are also reactive with tumors of extrapulmonary origin (Cuttita et al, 1981;Brown and Moore, 1982;Mulshine et al, 1983;Varki et al, 1984).…”
