Molecularly Imprinted Aptamers of Gold Nanoparticles for the Enzymatic Inhibition and Detection of Thrombin

Abstract: We prepared thrombin-binding aptamer-conjugated gold nanoparticles (TBA-Au NPs) through a molecularly imprinted (MP) approach, which provide highly efficient inhibition activity toward the polymerization of fibrinogen. Au NPs (diameter, 13 nm), 15-mer thrombin-binding aptamer (TBA(15)) with different thymidine linkers, and 29-mer thrombin-binding aptamer (TBA(29)) with different thymidine linkers (Tn) in the presence of thrombin (Thr) as a template were used to prepare MP-Thr-TBA(15)/TBA(29)-Tn-Au NPs. Thrombi… Show more

“…[12, 13, 15-18] Although gold nanoparticles may enable LSPR spectroscopy and improve sensitivity, they have so far been used as a layer underneath or on top of a molecularly imprinted polymer (MIP) film. [13, 17] In these configurations, nanoparticles are not used as direct transduction elements but for enhancing Raman scattering from analyte molecules (SERS) or propagating SPR on planar gold surface.…”

“…Nevertheless, it provides an alternative method to create aptamer‐MIPs. Aside from these copolymerization approaches, imprinting with thiol‐modified aptamers on the surfaces of gold nanoparticles for thrombin binding, was also reported …”

“…It is also possible to target both exosites 1 and 2 by directly coupling the mixture of TBA 15 and TBA 29 on the surface of AuNPs for thrombin inhibition . In addition, by tuning the thymine linker length (T n , n =0–60) and stem pair number (P m , m =0–16) on the terminal end of TBAs, Hsu et al.…”
