Molecular typing of Yersinia pseudotuberculosis by using an IS200-like element

Abstract: The IS200-like insertion sequence (IS) is a 708-bp element recently found in Yersinia pestis. Its nucleotide sequence is 85% identical to that of IS200 recovered in most Salmonella enterica isolates. It is also present in multiple copies in Y. pseudotuberculosis. In contrast, this IS is found in some (biotype 1B strains) but not other Y. enterocolitica strains and is absent in the nonpathogenic yersiniae: Y. frederiksenii, Y. kristensenii, Y. intermedia, Y. bercovieri, and Y. mollaretii. The number and locatio… Show more

“…In a previous study, we showed that HincII-restricted DNA from Y. pseudotuberculosis strain IP2777 (5.8, 4.3, 2.9, 2.6, 2.3, 2.0, 1.8 and 1 kb) and Y. enterocolitica strain 8081 (3.1 and 2.3 kb) contained sequences homologous to IS1541 [2]. We assumed that each hybridizing band harbored the unrestricted IS sequence since there was no HincII restriction site within IS1541.…”

“…Y. pseudotuberculosis strain IP2777 [2], Y. enterocolitica O:8 strain 8081 [2] and the cloning host strain DH5K of Escherichia coli were used in this study. Yersinia DNA was inserted into vector pUC19 (Appligene, Illkirch, France).…”

“…With time, IS elements can shift locations, replicate into other genomic positions, and can be excised from the genome. Collectively, we refer to these changes in the number and positions of IS elements IS200-like element [33] as transposition events. Polymorphism among strains results from variation in the number of elements, or the copy number, and their positions in the genome of the pathogen.…”

“…IS200 has been extensively used for the typing of Salmonella isolates of numerous serotypes by RFLP analysis (8,14,24), an approach that could be called IS200 typing (9). Similarly, molecular typing by RFLP analysis for IS1541 of isolates of Yersinia pseudotuberculosis has been described (29).…”
