Molecular Typing of Vibrio parahaemolyticus Isolates, Obtained from Patients Involved in Food Poisoning Outbreaks in Taiwan, by Random Amplified Polymorphic DNA Analysis

Abstract: Vibrio parahaemolyticus is one of the most important food-borne pathogens in Taiwan, Japan, and other countries with long coastlines. This paper reports on the development of a new random amplified polymorphic DNA (RAPD) method for the molecular typing of this pathogen. The 10-mer primer 284 (5′-CAG GCG CAC A-3′) was selected to generate polymorphic amplification profiles of the genomic DNA at an annealing temperature of 38°C. A total of 308 clinical isolates of V. parahaemolyticus collected during food poison… Show more

“…Due to considerable sequence similarity among the 16S rRNA gene sequences, an alternate method was employed to investigate the genetic diversity among the Pseudovibrio isolates. Using RAPD, which is frequently used to determine the genetic relationship between closely related bacterial species (Wong et al 1999;De Cesare et al 2001;Sudheesh et al 2002), 33 different RAPD types were identified among the 73 Pseudovibrio isolates, indicating that there is a genetically diverse population of Pseudovibrio spp. present which was not reflected in the 16S rRNA gene sequence analysis.…”

“…Unlike PCR, RAPD does not require a known target sequence for its operation. A 10-mer primer (5¢-CAG GCG CAC A-3¢) was selected to generate polymorphic amplification profiles of V. parahaemolyticus genomic DNA of 308 clinical and environmental isolates of V. parahaemolyticus by the RAPD method, generating 41 patterns that were grouped into 16 types (A-P) (Wong et al, 1999c). Several groups have used a variation of RAPD known as arbitrarily primed PCR to type urease-positive strains of V. parahaemolyticus (Okuda et al, 1997a) and the pandemic O3:K6 strains (Okuda et al, 1997b).…”
