Molecular structure of dynein and motility of a giant sperm axoneme provided with only the outer dynein arm

Abstract: The peculiar sperm axoneme of the dipteran Asphondylia ruebsaameni is characterized by an extraordinarily high number of microtubule doublets (up to 2,500) arranged in double parallel spirals. Doublets of the inner row of each spiral are tilted, so that their outer arms point towards the B-tubule of the next doublet in the outer row. Doublets are provided with only the outer arm, and no structure related to the central pair/radial spoke complex is present. When analyzed by quick-freeze, deep-etch electron micr… Show more

“…In nature, there are functionally motile flagella that have no CP or spoke apparatus (Woolley, 1998;Mencarelli et al, 2001). As discussed above, Chlamydomonas pf mutants, which lack the spokes and/or CP, are motile if they also carry a suppressor mutation (Brokaw et al, 1982;Huang et al, 1982;Porter et al, 1992).…”

“…The structural organization of this axoneme and the underlying morphogenetic processes have already been described, along with its motility properties [Mencarelli et al, 2000b[Mencarelli et al, , 2001. With respect to the 9ϩ2 axoneme, this model is characterized both by alterations in the assembly process of the axoneme structure and by the presence of a "simplified" motility mechanism.…”

“…Cilia/flagella beating is driven by outer arm dynein (OAD) and inner arm dyneins. The arrangement of dyneins on the axoneme has an overall periodicity of 96 nm, within which OAD binds every 24 nm; this 24-nm periodicity is completely conserved in essentially all eukaryotic organisms with "9 + 2" axonemes (2, 3) and even occurs in insect sperm flagella containing multiple rows of doublet microtubules arranged in a spiral configuration (4,5). OAD is best characterized in Chlamydomonas.…”
