Molecular structure, dynamic expression, and promoter analysis of zebrafish (Danio rerio)myf-5 gene

Abstract: We isolated a 1,438 bp cDNA fragment that encoded Myf-5 myogenic factor of zebrafish. The deduced amino acid contained 237 residues, including the basic helix-loop-helix domain that is conserved in all known Myf-5. The zebrafish myf-5 transcripts were first detectable at 7.5 hpf, increased substantially until 16 hpf, and then declined gradually to an undetectable level by 26 hpf. During somitogenesis, zebrafish myf-5 transcripts were distributed mainly in the somites and segmental plates. Prominent signals occ… Show more

“…It is preferable to conduct functional analysis of the regulatory elements of newly isolated genes in vivo rather than in vitro. In zebrafish, transgenes containing an endogenous promoter and the green fluorescent protein (GFP) gene have been used to map the cisacting elements of the promoter (Chen et al, 2001;Drivenes et al, 2000;Meng et al, 1997Meng et al, , 1999. They also have been used to target gene expression in specific tissues (Higashijima et al, 1997(Higashijima et al, , 2000Ju et al, 1999;Long et al, 1997).…”

“…Adaxial cells have a distinct cuboidal morphology and express the myogenic basic helix-loop-helix transcription factors myf5 and myod, followed by slow myosin heavy chain (MyHC) (Chen et al, 2001;Coutelle et al, 2001;Devoto et al, 1996;Weinberg et al, 1996). Subsequently, as somite borders form, myf5 and myod mark a distinct population of cells in the lateral somite (Chen et al, 2001;Coutelle et al, 2001;Weinberg et al, 1996).…”

The fate of the medial and lateral myogenic cells is known: medial adaxial cells form slow muscle and most migrate laterally to generate the superficial slow muscle fibres that lie within the somite under the In tetrapod phylogeny, the dramatic modifications of the trunk have received less attention than the more obvious evolution of limbs.…”

“…To study the regulation of AmphiMyoD gene expression and to determine if AmphiMyoD promoter could direct gene expression in muscle cells, we used zebrafish embryos as an alternative system to test if the AmphiMyoD promoter could direct expression of reporter gene, such as GFP, in muscle cells. Zebrafish embryos have been used successfully as an in vivo assay system for analysing the regulation of muscle specific gene expression (Chen et al 2001;Tan & Du 2002;Du et al 2003).…”
