The long-term usage of antibiotics has resulted in the evolution of multidrug-resistant bacteria. Unlike antibiotics, anti-virulence approaches target bacterial virulence without affecting cell viability, which may be less prone to develop drug resistance. Staphylococcus aureus is a major human pathogen that produces diverse virulence factors, such as α-toxin, which is hemolytic. Also, biofilm formation of S. aureus is one of the mechanisms of its drug resistance. In this study, anti-biofilm screening of 83 essential oils showed that black pepper, cananga, and myrrh oils and their common constituent cis-nerolidol at 0.01 % markedly inhibited S. aureus biofilm formation. Furthermore, the three essential oils and cis-nerolidol at below 0.005 % almost abolished the hemolytic activity of S. aureus. Transcriptional analyses showed that black pepper oil down-regulated the expressions of the α-toxin gene (hla), the nuclease genes, and the regulatory genes. In addition, black pepper, cananga, and myrrh oils and cis-nerolidol attenuated S. aureus virulence in the nematode Caenorhabditis elegans. This study is one of the most extensive on anti-virulence screening using diverse essential oils and provides comprehensive data on the subject. This finding implies other beneficial effects of essential oils and suggests that black pepper, cananga, and myrrh oils have potential use as anti-virulence strategies against persistent S. aureus infections.

Current and primary treatment modality in overactive bladder includes the administration of anticholinergics. The demand for new agents has been rising since anticholinergics have proven to come with many side effects. This study was designed to investigate the effects of ylang-ylang essential oil (YYEO) on the relaxation of urinary bladder muscle in vitro and in vivo. Effects of YYEO were assessed on resting tension, and electrical field stimulation- and various drug-induced contraction in vitro by checking the isometric tension changes of muscle strips and same procedures were repeated in the presence of methylene blue, Nw-Nitro-L-arginine methyl ester hydrochloride (L-NAME), or N-ethylmaleimide, and in vivo. YYEO decreased significantly the contractility of strips. There was no statistically significant difference between the treated group only with YYEO and the pretreated group with YYEO and methylene blue or L-NAME. When N-ethylmaleimide was employed, there was a statistically significant decrease in the rate of contraction. In vivo studies showed the same results compared with in vitro study. The results of this study indicate that YYEO has a relaxing effect on the bladder, and such mechanism is thought to be brought about by a pathway mediated by c-AMP.

Use of Topological Indices of Organic Sulfur Compounds in Quantitative Structure‐Retention Relationship Study

Structure-gas chromatographic retention index models were developed for some organic sulfur compounds at four different temperatures (60, 80, 100 and 120 8C) using only topological descriptors. At first, regression models were generated for each temperature separately with high values of multiple correlation coefficient and Fisher-ratio statistics. The results of cross validation test using leave-one-out (Q 2 ! 0.956) and leave-two-out (Q 2 ! 0.953) methods showed good predictive ability of the models developed. Then, a single combined quantitative structure-retention relationship model, added temperature as a parameter, was also developed for all the temperatures, showing good statistical parameters (R ¼ 0.991 and F ¼ 728.474). The stability and validity of the combined model were verified by both internal (Q 2 > 0.970) and external validation (Q 2 prediction set ¼ 0.993) techniques. The results of the study indicated the efficiency of the classical topological descriptors in simultaneous prediction of retention index values of sulfur compounds at different temperatures. The topological descriptors well covered the molecular properties known to be relevant for gas chromatographic retention data, such as molecular size and degree of branching.
