Molecular Signatures of Mitochondrial Complexes Involved in Alzheimer’s Disease via Oxidative Phosphorylation and Retrograde Endocannabinoid Signaling Pathways

Abstract: Objective. The inability to intervene in Alzheimer’s disease (AD) forces the search for promising gene-targeted therapies. This study was aimed at exploring molecular signatures and mechanistic pathways to improve the diagnosis and treatment of AD. Methods. Microarray datasets were collected to filter differentially expressed genes (DEGs) between AD and nondementia controls. Weight gene correlation network analysis (WGCNA) was employed to analyze the correlation of coexpression modules with AD phenotype. A glo… Show more

“…Endocannabinoids play a significant neuroprotective role in the AD model by regulating the timing of excitatory and inhibitory synaptic neurotransmitter release. They bind to and stimulate the cannabinoid 1 receptor (CB1R), thereby promoting synaptic plasticity and efficient neurotransmitter transmission, crucial for cognitive functions, memory, emotions, and motor skills (Abyadeh, Gupta, Paulo, et al, 2021; F. Chen et al, 2022). It’s important to note that mitochondrial transfer/transplantation has implications for tumorigenesis, drug resistance, and chemotherapy efficacy in cancer treatment.…”

“…Among other predicted targets, some of them also showed significant correlation with AD, such as FAAH (Fatty-acid amide hydrolase 1), SHBG (Sex hormone-binding globulin), and NOS2 (Nitric oxide synthase, inducible), indicating the complexity of anti-AD mechanisms of AA13. Specifically, multiple pieces of evidence illustrated that the retrograde endocannabinoid signaling pathway to which FAAH belongs is closely related to the mechanism of AD . MCHR1 (Melanin-concentrating hormone receptor 1) and HRH3 (Histamine H3 receptor) are important targets in the pathway of Neuroactive ligand–receptor interaction, which is also AD-related pathway .…”

“…Protein interaction analysis identified Uqcrfs1, Tufm, and Rpl4 as possible biological targets related to the evaluation of air quality. Uqcrfs1 is a core component of the mitochondrial respiratory chain ( 30 , 31 ). It transmits electrons to cytochrome C. Elevation of Uqcrfs1 expression leads to increased levels of cytochrome C1, resulting in volumetric hyperglycemia and metabolic abnormalities ( 32 ).…”
