Molecular recognition of glyconanoparticles by RCA and E. coli K88 - designing transports for targeted therapy

Abstract: The targeted drug delivery has been studied as one of the main methods in medicine to ensure successful treatments of diseases. Pharmaceutical sciences are using micro or nano carriers to obtain a controlled delivery of drugs, able to selectively interact with pathogens, cells or tissues. In this work, we modified bovine serum albumin (BSA) with lactose, obtaining a neoglycan (BSA-Lac). Subsequently, we synthesized glyconanoparticles (NPBSA-Lac) with the premise that it would be recognized by microbial galacto… Show more

“…The presence of a new band was observed as a signicant change in the spectrum of the modi-ed GO (GO-A and GO-AL) at 901 cm À1 , corresponding to Si-O and amino groups, attributed to the APTES conjugation. 18 The characteristic infrared bands of carbohydrates are localized between 900 and 1199 cm À1 ; 19 nevertheless, these were shied and referred to the SiO-X region. 18 We observed slight differences in this region for both GO-A and GO-AL, which made it difficult to identify the lactose groups.…”

“…Polymers, such as albumin, PLGA (poly(lactic-co-glycolic acid)), alginate, and chitosan have been widely used as vehicles for drug transportation to different sites within an organism (Goycoolea et al, 2009;Li et al, 2018;Sarmento et al, 2006;Gallegos-Tabanico et al, 2017;Danhier et al, 2012;Mir et al, 2017), making them encouraging tools for drug delivery. Polymeric nanoparticles (NPs) are of particular interest due to their properties, such as the simplicity of the synthesis method, biocompatibility, and biodegradability (Calvo et al, 1997;Goycoolea et al, 2009;Soares et al, 2018;Carreño-Gómez & Duncan, 1997).…”

“…Lactose was coupled to the amino groups of lysine residues present in the BSA, while galactose remained unchanged. Using this method, we obtained lactosylated bovine serum albumin (BSA-Lac) as the product [15,16]. Lactose has several advantages; it lacks immunogenicity, it is highly stable, and is easy to use for modification.…”
