Molecular phylogeny of the fungi of the Iceman's grass clothing

Abstract: To investigate the origin of the fungal hyphae that cover the grass clothing (cloak, boots) found near the neolithic mummy known as the Tyrolean Iceman, two radiocarbon-dated samples of grass were submitted to DNA extraction. The DNA was then PCR amplified using, respectively, primers specific for the region containing the internal transcribed spacers and the 5.8s rDNA (ITS), and primers specific for an approximately 600-bp long fragment of the nuclear small-subunit ribosomal DNA (SSU rDNA) repeat units of euk… Show more

“…These bands were purified from the gels and sequenced. A BLAST search (Altschul et al 1990) of GenBank using the 540-bp sequence revealed a close match with that of an ITS1-5.8S rRNA-ITS2 sequence from fungal clone T2709 (Rollo et al 1995) recovered from the clothing of The Iceman ''Ö tzi'' discovered near the Hauslabjoch formation in the Ö tztal Alps in 1991, and dated to 3350-3300 B.C. The alignment between T2709 (GenBank accession number X88771) and our Abydos 540-bp product (Ab540) is shown in Fig.…”

“…Molecular techniques are commonly used to solve problems in fungal taxonomy (Takamatsu, 1998 ;Ranghoo et al, 1999) and in several recent studies have also been used in the identification of fungi (Rollo et al, 1995 ;Ma et al, 1997 ;Zhang et al, 1997). The basic aim of the present study is to identify as many endophytes as possible from a limited number of palm fronds using traditional morphological techniques and molecular techniques.…”
