“…Our previous findings showed that 14-3-3γ could positively affect mTOR pathway (Yu et al 2014), which is essential for milk protein synthesis in dairy cow (Bionaz and Loor 2011;Zhang et al 2014). Further study on the immunoprecipitation of 14-3-3γ with 44 proteins in bovine mammary epithelial cells (BMECs) revealed that 14-3-3γ could interact with and positively affect the expression of eIF1AX (eukaryotic translation initiation factor 1A, X-linked) and RPS7 (ribosomal protein S7) to enhance milk protein synthesis (Yu et al 2014). These results suggest that 14-3-3γ is an important signaling molecule for milk protein synthesis through interacting with and regulating eIF1AX and RPS7 in the translational level.…”

“…The cell purity and milk protein synthesis ability of the primary BMECs were evaluated by the immunofluorescence detection of the expression of cytokeratin 18 and β-casein using methods as reported (Zavizion et al 1996;Yu et al 2014), and the purified BMECs in 0-10 passages were used for cell culture experiments.…”

“…Western blot analysis was performed as previously described (C. Yu et al, 2014). Briefly, 20 μg of protein equivalents of whole-cell extracts were extracted by WB Super RIPA Lysis Buffer (C2501;…”

“…Recent studies in bovines have highlighted a role of mTOR in the regulation of milk synthesis (Zhang et al, 2014), including milk protein synthesis (Wang et al, 2014b; Yu et al, 2014) and triacylglycerol or FAs synthesis (Li et al, 2014). Compared with untreated control cells, rapamycin reduced triglyceride secretion of BMECs (Zhang et al, 2014).…”
