Molecular Mechanisms of Lithium Action: Switching the Light on Multiple Targets for Dementia Using Animal Models

Abstract: Lithium has long been used for the treatment of psychiatric disorders, due to its robust beneficial effect as a mood stabilizing drug. Lithium’s effectiveness for improving neurological function is therefore well-described, stimulating the investigation of its potential use in several neurodegenerative conditions including Alzheimer’s (AD), Parkinson’s (PD) and Huntington’s (HD) diseases. A narrow therapeutic window for these effects, however, has led to concerted efforts to understand the molecular mechanisms… Show more

“…Thus, GSK3 inhibition generally appears to support the resolution of inflammatory processes. Consequently, the application of GSK3 inhibitors (e.g., SB216763, SB415286, TDZD-8 [274], indirubin-3 -monoxime [275], or lithium [145,276]) alleviates symptoms of systemic inflammation in murine and rat inflammatory disease models [121] (see Section 3.1.2) and equivalent results have been obtained in human cell culture approaches [223].…”

“…Although lithium’s mechanism of action remains incompletely understood, inhibition of IMPA and GSK-3, are considered to be central to its mood-stabilizing and neuroprotective effects ( 165 , 166 ). Inhibition of GSK-3 impacts several different signaling pathways implicated in neuronal function that also regulate TERT expression including the Wnt/beta-catenin pathway and the nuclear factor (erythroid-derived 2)-like 2 (Nrf2) pathway ( 167 , 168 ).…”

Moreover, lithium has been shown to increase CREB-dependent transcription of brain-derived neurotrophic factor (BDNF), which also upregulates TERT ( 166 , 169 ).…”

“…Определено несколько целей для разработки лекарств на основе лития: нейрогенное, цитопротективное, антиоксидантное, противовоспалительное действие, улучшение синаптической передачи. Однако для сравнительного анализа будет важно определить, какие из этих защитных механизмов наиболее необходимы для замедления когнитивного дефицита при деменции, и может ли комбинированная терапия объединить их для использования лития как нейропротекторного средства, избегая токсичности [33].…”
