Molecular markers for cell types of the inner ear and candidate genes for hearing disorders

Abstract: To identify genes expressed in the vertebrate inner ear, we have established an assay that allows rapid analysis of the differential expression pattern of mRNAs derived from an auditory epithelium-specific cDNA library. We performed subtractive hybridization to create an enriched probe, which then was used to screen the cDNA library. After digoxigenin-labeled antisense cRNAs had been transcribed from hybridization-positive clones, we conducted in situ hybridization on slides bearing cryosections of late embryo… Show more

“…Homogenin is a member of the villin/gelsolin family, a calcium-regulated, actin-modulating protein (Heller et al, 1998) and its downregulation would be in line with the previously described gelsolin downregulation during erythropoiesis (Hinssen et al, 1987). This critical role for cytoskeleton modification during erythroid differentiation is reinforced by our discovery of the Myosin alkali light chain downregulation.…”

“…The identities and integrity of the cDNA inserts were confirmed by sequencing. For screening, we first transformed yeast strain AH109 (Clontech) with each of the bait vectors, and then we individually transformed each bait-containing yeast cell population with plasmid DNA prepared from a chicken basilar papilla cDNA library in the HybriZAP two-hybrid bacteriophage vector (19). Auto-activation in bait-containing yeast cells was suppressed by adding 5 mM of the competitive His3 protein inhibitor 3-amino-1,2,4-triazole to the dropout medium.…”

“…We undertook an organ-specific cDNA library approach to identify genes important for hearing, a method that has been used successfully to identify various genes including auditory genes (Hedrick et al 1984;Jones and Reed 1989;Gurish et al 1992;Cohen-Salmon et al 1997;Soto-Prior et al 1997;Heller et al 1998;Jacob et al 1998;Robertson et al 1998). To this end, we made a human fetal cochlear cDNA library (Robertson et al 1994) and have used two complementary methods to identify genes within the cochlear library.…”
