Molecular Mapping of YrSP and Its Relationship with Other Genes for Stripe Rust Resistance in Wheat Chromosome 2BL

Abstract: Stripe rust, caused by Puccinia striiformis f. sp. tritici, is an important disease of wheat worldwide. Resistance is the best way to control the disease. YrSP, a gene originally from 'Spaldings Prolific' wheat and providing resistance to a broad spectrum of races, is used for differentiating P. striiformis f. sp. tritici races but its chromosomal location is not clear. To map YrSP, a near-isogenic line (AvSYrSPNIL) was backcrossed to the recurrent parent, Avocet S. Genetic analysis of the BC7F1, BC8, BC7F2, a… Show more

“…We selected SNP markers by "linkage map walking" on the distal and proximal side of the target region, and the initial linkage map was constructed with eight SNPs and qualitative disease reaction data to map Yr5 on the chromosome arm 2BL in two RIL populations. Moreover, Yr53 is flanked by wmc149 and wmc441 (Xu et al, 2013) where IWA4866 and IWA6121 are located at the distal side of the gene (Feng et al, 2015). Yr7, Yr43, Yr44, Yr53, and YrSP are race-specific allstage resistance genes, in addition to Yr5, and are located on the long arm of chromosome 2B (Zhang et al, 2009;Cheng and Chen, 2010;Xu et al, 2013;Feng et al, 2015).…”

“…The KASP markers based on SNP loci IWA6121 and IWA4096 were identified as flanking markers for Yr5 in both RIL populations. Yr7, Yr43, Yr44, Yr53, and YrSP are race-specific allstage resistance genes, in addition to Yr5, and are located on the long arm of chromosome 2B (Zhang et al, 2009;Cheng and Chen, 2010;Xu et al, 2013;Feng et al, 2015). These Yr genes are mapped to a similar position as Yr5 on an integrated consensus map (Maccaferri et al, 2015), though they are either defeated by contemporary PNW races or are from exotic sources.…”

“…As Yr76 is race-specific, it should not be used alone. Therefore, combinations of this gene with other widely effective all-stage resistance genes such as Yr5, Yr15, Yr45, Yr53, Yr61, Yr64, Yr65, and YrSP (Chen et al 2003;Feng et al 2015;Li et al 2010;Murphy et al 2009;Xu et al 2013;Zhou et al 2014a), and/or with genes for HTAP resistance such as Yr36, Yr39, Yr52, Yr59, Yr62, and many temporarily named QTL (Carter et al 2009;Chen 2013;Chen 2007, 2009;Lu et al 2014;Ren et al 2012;Uauy et al 2005;Zhou et al 2014b) are recommended.…”

“…Further assessment on the presence or absence of this gene in Ethiopian durum wheat may help to decide whether or not to use it in wheat breeding program. YrSp, originally identified from ' Spaldings Prolific ' wheat provides resistance to a broad spectrum of Pst races and is mapped on 2BL [57]. Recently, Marchal et al [40] identified that YrSp is allelic to Yr5 and is a truncated version of Yr5 sharing 99.8% sequence in common.…”
