Molecular Karyotyping and Exome Analysis of Salt‐Tolerant Rice Mutant from Somaclonal Variation

Abstract: LPT123-TC171 is a salt-tolerant (ST) and drought-tolerant (DT) rice line that was selected from somaclonal variation of the original Leuang Pratew 123 (LPT123) rice cultivar. The objective of this study was to identify the changes in the rice genome that possibly lead to ST and/or DT characteristics. The genomes of LPT123 and LPT123-TC171 were comparatively studied at the four levels of whole chromosomes (chromosome structure including telomeres, transposable elements, and DNA sequence changes) by using next-g… Show more

“…In rice, two CrRLK1L homolog genes, OsCrRLK1L2 and OsCrRLK1L3 , were found to be antagonistically upregulated in a diurnal and nocturnal manner, respectively (Nguyen et al 2015 ). Another rice CrRLK1L homolog gene, OsCrRLK1L15 , may participate in the salt stress response (Udomchalothorn et al 2014 ). In strawberries, a genome-wide analysis indicated that malectin-like receptor kinases in this species are associated with fruit ripening and abiotic stress responses (Zhang et al 2016 ).…”

“…The same could be hypothesized also for the MYB LOC_Os03g22560, which is closely related to the C3HC4 type zinc finger LOC_Os10g39770 and the trehalose synthase LOC_Os01g53000. In addition, within cluster 2, cell wall biosynthetic enzymes and a FERONIA receptor kinases, belonging to a class of proteins regulating cell wall biosynthesis [ 42 ] as reported above, were tightly transcriptionally co-regulated with signal factors like STRUBBELIG–RECEPTOR, MAPK, CrRLK1L, GATA zinc finger and mps, involved in a number of processes like hormone signalling, biotic and abiotic stress response, homeostasis, development and protein-protein interactions [ 96 98 ]. This association might reveal hypothetical involvements of these regulatory genes also in the regulation of cell wall biosynthesis.…”

“…Each replicate contained four plants per cultivar or line. Rice seedlings were grown in a hydroponic system containing a Wagner and Poesh solution (Udomchalothorn et al., 2014). The Wagner and Poesh nutrient solution consists of the following: potassium nitrate (KNO 3 ), calcium sulfate (CaSO 4 ), magnesium sulfate heptahydrate (MgSO 4 .7H 2 O), triple superphosphate‐monocalcium phosphate Ca(H 2 PO 4 ) 2, ammonium nitrate (NH 4 ) 2 NO 3 , unilate (magnesium oxide‐3.97%; iron‐1.5%; manganese‐1.5%; copper‐0.5%, zinc‐0.5%; boron‐0.3% and molybdenum‐0.026%), and iron ethylenediaminetetraacetic acid.…”
