Molecular Interactions of GBF1 with HY5 and HYH Proteins during Light-mediated Seedling Development in Arabidopsis thaliana

Abstract: Background: Light is an important factor for plant growth and development. Results: GBF1 interacts with two other bZIP proteins, HY5 and HYH, in light signaling. Conclusion: This work demonstrates the functional interconnections of GBF1, HY5, and HYH. Significance: The knowledge gained in this study will help to understand light-controlled plant growth and development.

“…The expression of HY5 in the cam7 hy5 double mutant also supports this notion ( Figure 8D). HY5 is known to interact and form heterodimers with at least two other bZIP proteins, GBF1/ ZBF2 and HYH, during Arabidopsis seedling development (Holm et al, 2002;Singh et al, 2012). Therefore, higher levels of HY5 might also interact or form heterodimers with other regulatory proteins in the regulation of HY5 expression.…”

“…For this experiment, the full-length CAM7 coding sequence was fused to the N terminus of YFP in the pUC-SPYNE vector and the full-length HY5 coding sequence was fused to the C terminus of YFP in the pUC-SPYCE vector (Singh et al, 2012). Whereas empty vectors did not produce any YFP fluorescence (Figure 7E), the interaction of CAM7 and HY5 produced strong YFP fluorescence in the nucleus ( Figure 7F).…”

“…To our surprise, OsbZIP23 could directly bind to the promoters of eight other OsPP2Cs under drought stress conditions (Supplemental Table S2). However, only OsPP2C49 and OsPP2C51 were differentially regulated by OsbZIP23 in the RNA-seq results (Supplemental Table S2), which indicates that the regulation of different clade A PP2Cs may need different transcription factors such as NACs and MYCs to coordinate with bZIP transcription factors for the differential regulation (Singh et al, 2012;Xu et al, 2013;Maurya et al, 2015).…”

“…FHY3 binds to an FHY3 binding site and HY5 to an ACGT-containing element in the COP1 promoter, both of which seem to be required for the UV-B responsiveness of COP1 . Combinatorial regulation exerted by the interaction of other transcription factors with HY5/HYH is widely present, with negative or positive regulatory effects (Shin et al, 2007;Andronis et al, 2008;Holtan et al, 2011;Huang et al, 2012;Jiang et al, 2012;Singh et al, 2012;Gangappa et al, 2013aGangappa et al, , 2013bJing et al, 2013;Abbas et al, 2014). Moreover, HY5 interacts with the chromatin-remodeling factor PICKLE (PKL), recruiting PKL to promoters of target genes repressing the H3K27me3-repressive histone mark (Jing et al, 2013).…”
