Molecular Insights into X;BTA5 Chromosome Rearrangements in the Tribe Antilopini (Bovidae)

Abstract: For a clade that includes Antilope, Gazella,Nanger and Eudorcas (Antilopinae), X;BTA5 translocation is a synapomorphy. Using a combination of fluorescence in situ hybridization (FISH) probes and polymerase chain reaction techniques, we provide (i) the first insight into the X;BTA5 architecture which differs in the species under study: Antilope cervicapra (genus Antilope), Gazella leptoceros (genus Gazella) and Nanger dama ruficollis (genus Nanger), (ii) determination of interstitial satellite DNA at the X;BTA5… Show more

“…Blood samples were cultured, harvested and fixed according to standard protocols as described previously [ 35 ]. Metaphase chromosome spreads for laser microdissection as well as slides for FISH analysis were prepared according to the procedures described previously [ 36 ]. For the assessment of deer karyotypes, GTG-banding was performed using the standard trypsin/Giemsa procedure [ 37 ].…”

“…The analysis was performed following the protocols for FISH with painting or BAC probes. The posthybridization washing, image capture and processing were carried out as described in [ 36 ]. The BAC probes labelled with biotin-16-dUTP and digoxigenin-11-dUTP were detected with Avidin-CY3 (Amersham Pharmacia Biotech, Piscataway, NJ, USA) and antidigoxigenin-fluorescein (Roche).…”

“…Therefore, we cannot affirm that the amount of ITS observed is sufficient to isolate the autosomal and sexual segments, but it may contribute to the insulation buffer. These observations lead us to search for other repetitive sequences that could be colocated with ITSs and block the diffusion of X inactivation to the autosomal segment, such as rDNA [Dobigny et al, 2004;RodriguesNoronha et al, 2004;Cernohorska et al, 2012Cernohorska et al, , 2015. In this case, rDNA clusters have already been ruled out since they do not map to the centromeric region of Rb(X.1) in M. minutoides [Cazaux et al, 2011].…”

“…Interestingly, all species of Tragelaphini exhibit Y;BTA13 translocation, which would result in an imbalance in diploid numbers between sexes with the exception of T. spekii (2n = 30) and T. imberbis (2n = 38) [Rubes et al, 2008]. For a clade that includes Antilope, Gazella, Nanger and Eudorcas (Antilopinae), X;BTA5 translocation is a synapomorphy [Cernohorska et al, 2012]. Using region-specific painting probes, the authors revealed that tandem fusion (head-totail) assembled the 2 chromosomes in G. leptoceros and N. dama ruficollis , unlike centric fusion (head-to-head) detected in A. cervicapra .…”
