Molecular Insights into Epigenetics and Cannabinoid Receptors

Abstract: The actions of cannabis are mediated by G protein-coupled receptors that are part of an endogenous cannabinoid system (ECS). ECS consists of the naturally occurring ligands N-arachidonylethanolamine (anandamide) and 2-arachidonoylglycerol (2-AG), their biosynthetic and degradative enzymes, and the CB1 and CB2 cannabinoid receptors. Epigenetics are heritable changes that affect gene expression without changing the DNA sequence, transducing external stimuli in stable alterations of the DNA or chromatin structure… Show more

“…There is increasing recent concern at the epigenetic impacts and, therefore, transgenerational effects of many cannabinoids [ 54 , 95 , 96 ]. Recent serial whole epigenome screening has identified many sets of functional annotations identified in the Ingenuity Pathway Analysis of human sperm, including particularly the following, which are of relevance to the morphogenesis of facial structures [ 8 ]: Head development (47 genes, page 304, p = 0.00012).…”

“…Current research has only begun to uncover the pathways involved in cannabisinduced changes in DNA methylation, histone modifications, and miRNA expression [59]. Of the existing research, evidence supports the hypothesis that cannabis can alter the activity of histone and DNA-modifying enzymes [58].…”

“…In a review by Basavarajappa et al, several studies highlight the importance of further evaluation of CB receptor regulation by epigenetic modifications (13,44). Di Marzo et al eloquently describes the discovery of an expanded eCB, the endocannabinoidome (eCBome), an elaborate signaling system that participates in neuronal development and synaptic plasticity in most brain areas (13,(45)(46)(47) and is subsequently involved in the onset, progression, and symptomology of major neuropsychiatric disorders, providing a potential target for the development of novel therapeutics against these disorders [see Figure 2 (11)(12)(13)].…”
