Molecular Imaging of Tumors Using a Quantitative T1 Mapping Technique via Magnetic Resonance Imaging

Abstract: Magnetic resonance imaging (MRI) of glioblastoma multiforme (GBM) with molecular imaging agents would allow for the specific localization of brain tumors. Prior studies using T1-weighted MR imaging demonstrated that the SBK2-Tris-(Gd-DOTA)3 molecular imaging agent labeled heterotopic xenograft models of brain tumors more intensely than non-specific contrast agents using conventional T1-weighted imaging techniques. In this study, we used a dynamic quantitative T1 mapping strategy to more objectively compare int… Show more

“…84 A quantitative MRI study recently conducted demonstrates a sustained binding of SBK2 to tumors over time compared with conventional contrast agents with an approximate 2:1 TNR 45-60 minutes after injection. 85 Toxicity, dosage, and efficacy of the PTPμ-directed agents still needs to be tested in humans, but these preliminary studies are encouraging, especially considering the high level of tumor margin labeling. Human studies are being planned.…”

“…6 For this in vitro study, 100 signal averages were acquired for determination of the agent relaxivities ( r 1 ). The T 1 relaxivities were obtained by measuring a dilution series of two lots of each agent on three separate occasions using saline as the diluent.…”

“…47 Previous studies using heterotopic glioma xenograft models demonstrated that a PTPμ-targeted MRI agent with 3 Gd chelates labeled flank tumors more effectively than non-specific contrast agents. 5,6 The chelator used was the polydentate ligand 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA). A dynamic, quantitative T 1 mapping strategy was implemented to objectively evaluate the in vivo intra-tumoral concentration of the PTPμ-targeted contrast agent [SBK2-Tris-(Gd-DOTA) 3 ] over time.…”
