Molecular identification of the liver- and the heart-type fatty acid-binding proteins in human and rat kidney. Use of the reverse transcriptase polymerase chain reaction

Abstract: The cDNAs of two types of fatty acid-binding protein (FABP) present in human kidney, previously described as types A and B, were isolated using reverse transcriptase-PCR (RT-PCR) with human kidney mRNA and various sets of primers. The cDNA fragments were cloned and sequenced. Renal FABP type A and B cDNAs appeared to be completely identical to human liver- and heart-type FABP cDNAs respectively. In the second part of this study we demonstrated the presence of liver-type FABP in rat kidney by chromatography, e.… Show more

“…There was a discrepancy in previous studies on the localization of L-FABP in the mammalian kidney in that L-FABP was localized in PTC and DTC in the rat kidney, 14 but only in PTC in the human kidney. 13 We performed immunohistochemical analyses for the distribution of L-FABP in the rat kidney.…”

“…The finding is compatible with a previous report on the low mRNA expression of L-FABP both in the human and rat kidney. 14 In rats that had been treated with different doses of CCl 4 , the level of L-FABP mRNA expression in the kidney was low and did not differ from that in the kidney of control rats (Figure 2b). These results suggest that renal L-FABP synthesis is very low and that there may be a pathway to transport L-FABP to the kidney via the circulation.…”

“…4,5 L-FABP is expressed in the human proximal tubules of the kidney. 6,7 human L-FABP (hL-FABP) binds fatty acids and transports them to mitochondria or peroxisomes, where fatty acids are ␤-oxidized, and participates in intracellular fatty acid homeostasis. 8,9 Some experimental studies of animal models of kidney disease with tubulointerstitial damage revealed that renal hL-FABP was up-regulated and urinary excretion of hL-FABP was increased.…”

“…6 Liver-type fatty acid-binding protein (L-FABP) is a 14-kDa protein found in the cytoplasm of human PTCs. 7 Fatty acids are bound to L-FABP and transported to the mitochondria or peroxisomes, where they are β-oxidized. This reaction plays a role in fatty acid homeostasis.…”
