Leaf curl disease of tobacco (TbLCD) is endemic in India. A monopartite Begomovirus, a betasatellite and an alphasatellite were found associated with the disease in Pusa, Bihar. The DNA-A of the Begomovirus associated with TbLCD in Pusa, Bihar was found to comprise of 2707 nt with a typical Old World begomovirus-like genome organization. The full-length sequence of DNA-A [HQ180391] showed that the Pusa isolate is a newly described member of the genus Begomovirus, as it had <89% sequence homology with DNA-A of all the known begomoviruses. The isolate is tentatively named as Tobacco leaf curl Pusa virus [India:Pusa:2010]. The betasatellite (HQ180395) associated with TbLCD in Pusa was identified as a variant of Tomato leaf curl Bangladesh betasatellite [IN:Raj:03], with which it shared 90.4% sequence identity. The alphasatellite (HQ180392) associated with the disease had highest 87% nucleotide sequence identity with Tomato leaf curl alphasatellite. The Begomovirus, betasatellite, and alphasatellite associated with TbLCD in Pusa, Bihar, India were found to be recombinants of extant begomoviruses, betasatellites and alphasatellites spreading in the Indian sub-continent and South-East Asia.

Vorstenlanden tobacco is the best product of PT Perkebunan Nusantara X (PTPN X) Klaten, Central Java that is commonly produced into high economic value cigars. During the planting season of 2010/2011, there was an epidemy of leaf curl disease that caused billion rupiahs financial loss. Several efforts had been done, including the use of pesticides, but the result had not been satisfactory. Therefore, this research aimed to identify the pathogen of tobacco leaf curl disease and conduct integrated control system using three combinations in the nursery [A (biological agents treatment, plastic screenhouse, without physical barrier); B (biological agents treatment, plastic screenhouse + screen plot, with physical barrier of single screen, and sprayed insecticide); C (biological agents treatment, plastic screenhouse + single screen plot, with physical barrier of double screens, and fogging using white oil + insecticide)] and planting area [A (physical barrier of single screen and sprayed insecticide); B (physical barrier of single screen, sprayed insecticide, and sanitation of broad leaf weed + eradication of infected plant); C (physical barrier of double screens, fogging white oil + insecticide, and sanitation of broad leaf weed + eradication of infected plant)]. The result showed that leaf curl disease that occured at PTPN X Klaten was caused by Begomovirus, based on PCR result using Krusty and Hommer primers for Begomovirus Coat Protein gene. The most effective integrated disease control of leaf curl disease in the nursery was C combination, that consisted of biological agents treatment on the seedlings medium, screenhouse covered by plastic and double screens that was combined with physical barrier of double screens around the field, and fogging using white oil+insecticide of pyrethroid active ingredient. The most effective integrated disease control of leaf curl disease in the field was the same C combination, that consisted of the use of physical barrier of double screens, environmental sanitation of weeds around the field and eradication of infected plants and fogging using white oil + insecticide of pyretroid active agent.Keywords: Begomovirus, integrated disease control, leaf curl disease of tobacco, white oil INTISARI Tembakau vorstenlanden merupakan produk andalan PT Perkebunan Nusantara X (PTPN X) Klaten, Jawa Tengah sebagai bahan baku cerutu yang bernilai ekonomi tinggi. INTISARIPeningkatan populasi kutu kebul (B. tabaci Gen.) di daerah Klaten, Jawa Tengah pada tahun 2009 ternyata menjadi penyebab mewabahnya penyakit kerupuk pada tanaman Solanaceae di wilayah tersebut. Tanaman tembakau cerutu Vorstenlanden milik PTPN X (Persero) yang ditanam di area bawah naungan (TBN) yang berada di wilayah tersebut turut terjangkit wabah penyakit kerupuk setelah sebelumnya terindikasi terjadi peningkatan jumlah kutu kebul. Wabah penyakit kerupuk pada tembakau cerutu Vorstenlanden milik PTPN X (Persero) telah menurunkan hasil hingga 70%. Gejala penyakit pada tembakau berupa penyakit kerupuk. Penelitian ini bertujuan untuk mengetahui efektivitas penerapan barier fisik sebagai langkah awal untuk mengendalikan populasi kutu kebul pada pertanaman tembakau cerutu milik PTPN X (Persero). Penelitian dimulai dengan memonitoring populasi kutu kebul dan peningkatan intensitas penyakit kerupuk pada tanaman tembakau. Percobaan dilakukan secara demplot pada lahan pertanaman tembakau di wilayah PTPN X (Persero). Teknis budidaya tembakau seluruhnya mengikuti Standar Operasional Prosedur (SOP) dari PTPN X (Persero), sedangkan untuk strategi pengendalian kutu kebul dilaksanakan dengan mengikuti 2 macam perlakuan yaitu ; (1) penerapan sistem barier tunggal berupa kelambu (bermesh rapat) dan kelambu standar dari PTPN X sebagai barier fisik (2) penerapan sistem barier ganda yaitu kombinasi barier fisik dan barier biologis berupa tanaman jagung penghalang. Hasil menunjukkan bahwa penerapan sistem barier baik tunggal maupun ganda dapat menekan populasi kutu kebul sehingga berdampak pada penurunan intensitas penyakit kerupuk pada tembakau.
