Molecular Genetic Dissection of Spawning, Parentage, and Reproductive Tactics in a Population of Redbreast Sunfish, Lepomis auritus

Abstract: Despite a great diversity of reproductive behaviors in fishes, few studies have examined the genetic consequences of alternative reproductive tactics. Here we develop and employ microsatellite markers to assess genetic paternity and maternity of progeny cohorts in a population of redbreast sunfish (Lepomis auritus), a species in which males build and tend nests. Nearly 1000 progeny from 25 nests, plus nest-attendant males and nearby adults, were genotyped at microsatellite loci that displayed more than 18 alle… Show more

“…We did not find specimens of C. virginicus in areas with finer substrata associated with lower flows along the periphery of the stream. Although the one nest of Lepomis auritus sampled was constructed of gravel, it was located along the periphery of the stream, surrounded by finer substrata, and did not contain specimens of C. virginicus [nor did any of the 25 nests of L. auritus surveyed from Fourmile Creek in the Savannah River system by DeWoody et al (1998); (J.A. DeWoody, pers.…”

“…DNA was later isolated from the adults and a subset of larvae, and microsatellite DNA fingerprints were obtained by using multiplex genotyping (Colbourne et al, 1996;Neff et al, 2000). Up to four primer sets (locus: RB7, RB20, LMAR10, LMAR14; published in DeWoody et al, 1998 andSchable et al, 2002) were simultaneously amplified, and all competing parental and cuckolder offspring were unambiguously identified by using exclusion methods (Chakraborty et al, 1988).…”

“…Each individual was genotyped at three microsatellite loci: RB7, LMAR10 and either RB20 or LMAR14 (for details of these loci see [9,10]). RB20 was used for the parentage analysis of the 2005 bluegill samples, whereas LMAR14 was used for the interspecies allele frequency comparisons and the parentage analysis of the 2003 and 2010 pumpkinseed samples.…”
