Molecular Genetic Analysis of Sexual Rejection: Roles of Octopamine and Its Receptor OAMB inDrosophilaCourtship Conditioning

Abstract: After Drosophila males are rejected by mated females, their subsequent courtship is inhibited even when encountering virgin females. Molecular mechanisms underlying courtship conditioning in the CNS are unclear. In this study, we find that tyramine ␤ hydroxylase (T␤H) mutant males unable to synthesize octopamine (OA) showed impaired courtship conditioning, which could be rescued by transgenic T␤H expression in the CNS. Inactivation of octopaminergic neurons mimicked the T␤H mutant phenotype. Transient activati… Show more

“…Octopamine immunoreactivity is observed in the calyx and γ lobes (Sinakevitch and Strausfeld, 2006). While the αβ and α′β′ lobes are devoid of octopamine immunoreactivity, MARCM and GRASP analyses that allow high spatial resolution imaging reveal the octopamine neurons innervating the αβ lobes (Zhou et al, 2012). The calyx has OAMB-AS, but not OAMB-K3, immunoreactivity and this immunoreactivity is contributed by extrinsic, but not intrinsic, mushroom body neurons (J. Lim and K.-A.…”

“…Since octopamine is known to control aggression in Drosophila males (Baier et al 2002;Hoyer et al 2008;Zhou et al 2008Zhou et al , 2012Certel et al 2010;Erion et al 2012), to establish if TfAP-2 and Twz were required for octopamineregulated male behavior, we performed an aggression assay. To do this, Tdc2-GAL4, UAS-Tfap2 RNAi , UAS-Twz RNAi , and UASTfap2 OE were all crossed to w 1118 , and the heterozygous progeny were used as controls.…”

“…The behavioral decision to be aggressive is in part controlled by systems that also regulate metabolism, such as the monoamine system (Dierick and Greenspan 2007;Hoyer et al 2008;Zhou et al 2008;Alekseyenko et al 2010). In the fruit fly Drosophila melanogaster, it was determined that a major regulator of male aggressive and mating behavior is the noradrenaline analog, octopamine (Baier et al 2002;Hoyer et al 2008;Zhou et al 2008Zhou et al , 2012Certel et al 2010;Erion et al 2012). In addition, the monoamines dopamine and serotonin have been linked to the regulation of aggressive behavior (Lucki 1998;Baier et al 2002;Alekseyenko et al 2010Alekseyenko et al , 2013Belsare et al 2010).…”

“…sNPF is expressed in the mushroom bodies and has been implicated in regulating IPCs (Nassel et al 2013) while it also acts as a neuromodulator in olfactory memory (Knapek et al 2013). Mushroom bodies also play important roles in courtship behavior and courtship learning or conditioning (Sakai and Kitamoto 2006;Keleman et al 2012;Zhou et al 2012). Finally, the expression of Nep1 in abdominal ganglia suggests a possible role in modulating sperm transfer and copulation duration that is regulated by corazonin expressed in the abdominal ganglia (Tayler et al 2012).…”
