Inflammatoryboweldisease(IBD)isachronicinflammatoryconditionwithanunknownetiology.Nicorandil,apotassiumchannel opener, has been used for many years for the treatment of angina. Recently, it has been shown that nicorandil possesses some novel traits such as anti-apoptotic, gastroprotective, free radical scavenging, and anti-inflammatory properties. Therefore, we set out to examine the possible beneficial effect of nicorandil in a rat model of IBD. Colitis was induced by rectal administration of 2,4,6-trintrobenzene sulphonic acid (TNBS) into rats. Groups of animals used in this study were sham, control, and exposure to dexamethasone,nicorandil,glibenclamid(apureadenosinetriphosphatesensitivepotassiumchannel(KATP)blocker),ornicorandilplus glibenclamid.Drugswereadministeredbygavageandanimalsweresacrificedafter7days.Biochemicalmarkers,includingTNF-αand IL-1β,ferricreducing/antioxidantpower(FRAP),myeloperoxidase(MPO)activityandthiobarbitoricacid-reactivesubstance(TBARS), weremeasuredinthehomogenateofcolonictissue.Resultsindicatethatnicorandilsignificantlyreducesmacroscopicandhistological damageinducedbyTNBS.NicorandildiminishesMPOactivityandlevelsofTBARS,TNF-α,andIL-1βindamagedcolonictissuewith aconcomitantincreaseinFRAPvalue(P<0.01).Theseeffectswerenotreversedbycoadministrationofglibenclamide.Inconclusion, nicorandilisabletoameliorateexperimentalIBDwithadoseinwhichitdoesnotshowanyanti-hypertensiveeffect,andthemechanism ofwhichispartiallyortotallyindependentfromKATPchannels.Itishypothesizedthatnitricoxidedonationandfree-radicalscavenging propertiesofnicorandilupregulateendothelialnitricoxidesynthasemayberesponsibleforthisphenomenon.Thesefindingssuggest thatnicorandilcanbeusefulintreatmentofIBD,althoughfurtherinvestigationsareneededtoelucidatethemechanismsinvolved.

Inflammatory bowel disease (IBD) is a multifactorial disease with an unknown etiology characterized by oxidative stress, leukocyte infiltration and a rise in inflammatory cytokines. In this study, we have investigated the effects of silymarin, a mixture of several flavonolignans with established antioxidant and anti-inflammatory properties, on trinitrobenzene sulphonic acid (TNBS)-induced colitis in rats. Experimental colitis was induced in male Wistar-albino rats by delivering TNBS to the distal colon. All the medicines were administered by gavage for seven days. Thirty-six male rats were divided into six groups containing six rats in each one. Control rats received only TNBS. In the treated groups, animals were given different doses of silymarin (40, 80, and 160 mg/kg). Dexamethasone (1 mg/kg) was used as the positive treatment. Colonic status was investigated seven days post induction of colitis through macroscopic, histological, and biochemical analyses. Amelioration of the morphological signs including macroscopic damage, necrotic area, and histology were seen subsequent to treating animals with silymarin. These observations were accompanied by a significant reduction in the degree of both neutrophil infiltration, indicated by decreased myeloperoxidase activity, and lipid peroxidation, as measured by a decline in malodialdehyde content in inflamed colon as well as a decrease in levels of inflammatory cytokines (TNF-α and IL-1β). The results of the present study reveal that the beneficial effect of silymarin in bowel cells is mediated through its anti-oxidant and anti-inflammatory potentials.

Context: Tragopogon graminifolius DC. (Compositae) (TG) has been proposed as an efficacious remedy for gastrointestinal ulcers in Iranian traditional medicine. Objective: The present study evaluates the efficacy of TG on experimental colitis and the responsible mechanisms. Materials and methods: After induction of IBD by 2,4,6-trinitrobenzenesulfonic acid (TNBS), rats received standardized ethanol extract of TG aerial part at 20, 30, or 50 mg/kg/d orally. After 12 d, the rats were sacrificed and the colon was removed and assessed for macroscopic and microscopic changes. Also, tumor necrosis factor-alpha (TNF-a), interleukin-1 beta (IL-1b), total antioxidant capacity, myeloperoxidase (MPO), and lipid peroxidation (LPO) were measured in the colon homogenate. Result: TG extract significantly reduced macroscopic and microscopic scores of colitis with ED 50 values of 23 and 39 mg/kg, respectively. MPO was significantly reduced in all plant extract groups with an ED 50 value of 41 mg/kg. The ED 50 values of extract for inhibition of TNF-a and LPO were 44 and 93 mg/kg, respectively. IL-1b significantly decreased by 50 mg/kg of TG extract (ED 50 ¼ 57 mg/kg). Total antioxidant power markedly increased by 50 mg/kg group (ED 50 ¼ 43 mg/kg). Discussion: TG exhibited efficacy on TNBS-induced colitis via anti-inflammatory, immunomodulatory, antioxidant, and mucosal healing properties. Conclusion: TG possesses promising healing function on colitis. Clinical trials are warranted to prove its efficacy and tolerability in IBD.
