Molecular epidemiology of tetM genes in Neisseria gonorrhoeae

Abstract: Objective: To examine the epidemiology of the tetM gene in Neisseria gonorrhoeae strains with high level resistance to tetracycline (TRNG) using a polymerase chain reaction (PCR) assay. Methods: A single tube PCR was developed which distinguishes between the American and Dutch variants of the tetM gene. Between 1988 and 1995, 518 strains of TRNG (tetracycline MIC >8 mg/l) were referred to the Gonococcus Reference Unit by other laboratories or isolated from routine swabs taken at local clinics. The strains were… Show more

“…The tetM resistant determinant, which confers high-level resistance to tetracycline, is borne on plasmids and is transferred either through conjugation or transformation (Knapp, Johnson, Zenilman, Roberts, & Morse, 1988;Morse, Johnson, Biddle, & Roberts, 1986;Turner, Gough, & Leeming, 1999). It was found in only 5 of the 10 strains with high-level resistance to tetracyline (MIC equal or greater than 16 µg/ml).…”

“…Isolates were tested for ␤-lactamase production by the chromogenic cephalosporin method. PCR detection and characterization of the tetM gene were performed on isolates of N. gonorrhoeae by using a one-step PCR amplification (13). PCR products were electrophoresed in a 1% agarose gel containing ethidium bromide and visualized by UV fluorescence.…”

“…The high level of tetracycline resistance (MIC of 24 g/ml) was due to a plasmid defined as an "American type plasmid," the most frequently detected plasmid in tetracycline-resistant N. gonorrhoeae isolates (16).…”

“…The tetracycline resistance mobile elements were detected by PCR using three primers, UF (5Ј-CTCGAACAAGAGGA AAGC), AR (5Ј-GCATTCCACTTCCCAAC), and DR (5Ј-T GCAGCAGAGGGAGG), as previously described (10,11,16). The high level of tetracycline resistance (MIC of 24 g/ml) was due to a plasmid defined as an "American type plasmid," the most frequently detected plasmid in tetracycline-resistant N. gonorrhoeae isolates (16).…”
