Aim:Anaplasma marginale is a rickettsial pathogen responsible for progressive anemia in ruminants leading to huge economic losses. The objectives of this cross-sectional study were to determine the prevalence of anaplasmosis and therapeutic evaluation of traditional line of treatment.Materials and Methods:A total of 168 cattle presented to Teaching Veterinary Clinical Complex, Lala Lajpat Rai University of Veterinary & Animal Sciences, Hisar during the period of 3 months (July-September, 2014) with history of fever, anorexia, reduced milk yield and tick infestation were analyzed for prevalence of hemoprotozoan diseases using classical giemsa stained thin blood smear parasitological method.Results:Out of these 168 animals, 7 (4.17%) were found to be suffering from anaplasmosis on the basis of presence of dense, rounded, intra-erythrocytic bodies situated on or near the margin of the erythrocytes. Overall prevalence of theileriosis and babesiosis were found to be 42.9% and 1.8%, respectively. Level of parasitemia was noticed to be 1.2%, 0.8% and 0.9% in babesiosis, theileriosis, and anaplasmosis, respectively. The most marked and common clinical signs reported in all the cases were severe anemia (hemoglobin=3-6 g/dl) and history of fever, followed by normal body temperature. Following treatment with oxytetracycline parenterally along with supportive therapy out of seven cases six got recovered without any side-effects.Conclusion:The current study indicates the emerging status of anaplasmosis in this part of the country as during the past few years there are very few reports showing the prevalence of clinical cases of anaplasmosis. Treatment with oxytetracycline yielded excellent result showing recovery in most of the clinical cases.

Data on the implication of the stable fly (Stomoxys calcitrans, Diptera: Muscidae), in the transmission of Anaplasma marginale in tick-free cattle is scarce. Hence, the objective of this investigation was to detect the presence of DNA from A. marginale in stable flies caught near a bovine herd, which has been maintained free of ticks for 40 years, and occasionally presents some clinical cases of anaplasmosis. Twenty-four batches of S. calcitrans (15 flies each) were collected in the morning and in the afternoon twice a week (except for one catch in one week of September and one catch in one week of December) during 12 catching days in a period of four months. Their DNA was obtained and analysed by nested PCR (nPCR) to identify the presence of A. marginale DNA. Seven of the batches (29.16 %) were positive for A. marginale, as detected by an nPCR that targets the A. marginale msp5 gene. Similarly, in two out of 12 catching days (16.66 %), those S. calcitrans batches collected in the morning and in the afternoon, were positive for A. marginale, while those S. calcitrans groups collected in three out of 12 catching days (25 %), were positive for the rickettsia. The obtained results suggest that A. marginale transmission is carried out mechanically by S. calcitrans, favouring the circulation and maintenance of the microorganism in this particular bovine herd.

The current description of biological transmission of Anaplasma marginale by Rhipicephalus microplus ticks, includes of the biological intrastadial and transstadial transmission. Both transovarian transmission of Anaplasma from engorged ticks to their progeny and, transmission from infected unfed larvae to the mammalian host is controversial. In order to demonstrate vertical transmission of A. marginale by R. microplus ticks under experimental conditions, feed-acquisition infected engorged females were incubated at 18 °C or 28 °C for oviposition. Larvae hatched from these ticks were used to infest two steers for each incubation temperature. None of the four steers infested with either lot of larvae developed clinical disease, yet subclinical infection was observed in the steers infested with larvae from engorged ticks incubated at 28 °C for hatching. gDNA from, larvae used for the infection of the carrier tick donor, gDNA from larvae oviposited at 28 ºC, gDNA from blood of A. marginale-positive steers, were positive for amplification of msp5 and msp1α the variable region by PCR. All other DNA samples from the original stabilate, blood from the donor steer, larvae from ticks incubated at 28 °C and blood from steers infested with these same larvae were positive to both, msp5 and msp1α PCR. msp1α sequences of all PCR products were the same and are consistent with previously reported Tlapacoyan-2 sequence. The present evidence indicates that R. microplus is capable of passing A. marginale to its progeny and that these infected larvae can transmit the infection to susceptible hosts.
