Molecular dynamics insights into the structure, function, and substrate binding mechanism of mucin desulfating sulfatase of gut microbe Bacteroides fragilis

Abstract: The complex and dynamic consortia of microbiota that harbors the human gastrointestinal tract contributes ominously to the maintenance of health, the onset and progression of diverse spectrum of disorders. The capability of these enteric microbes to bloom within the gut mucosal milieu is often associated to the glycan metabolism of mucin-degrading bacteria. Accruing evidences suggests that the desulfation of mucin is a rate-limiting step in mucin degradation mechanism by colonic bacterial mucin-desulfating sul… Show more

“…We looked broadly at reports of SCFA production among the reviewed case-control studies and found that in general the genera reduced in MDD have extensive capacity to metabolize carbohydrates, particularly mono- and disaccharides and their derivatives [ Bifidobacterium (5862), Faecalibacterium (63), and Ruminococcus (6466)]. Bacteroides , found to be reduced in a treatment-nonresponsive depressed subgroup compared to healthy controls (27), as a genus has a particularly rich armamentarium for metabolizing more complex carbohydrates including glycans of human mucin (6778).…”

“…thetaiotaomicron, Bacteroides ovatus, and Prevotella sp. strain RS2 Bifidobacterium breve UCC2003, or B. fragilis possess mucin-desulfating sulfatases or glycosulfatases (Salyers et al, 1977, Berteau et al, 2006, Benjdia et al, 2011, Egan et al, 2016, Praharaj et al, 2018.…”
