Molecular Distribution of IGF Binding Protein-5 in Human Serum

Abstract: IGF binding protein-5 (IGFBP-5) forms ternary complexes with IGFs and the acid-labile subunit (ALS) in vitro, but these complexes have not been demonstrated in the circulation. To examine the molecular distribution of circulating IGFBP-5 we developed an RIA with high specificity for IGFBP-5 among the IGFBPs, but wide cross-reactivity among primate and nonprimate species. The mean serum IGFBP-5 level (+/-SD) was 208 +/- 73 ng/ml in healthy men and 206 +/- 67 ng/ml in nonpregnant women, decreasing to 114 +/- 38 … Show more

“…3A, bottom panel, lane 2). As noted previously (16), the relative intensity of the IGFBP-3 bands is reduced in these mice, probably because IGFBP-5 may sequester IGF-I from IGFBP-3, leading to accelerated degradation of IGFBP-3 (7). In marked contrast to this pattern, no band equivalent to wtBP5-myc was observed in the ligand blot for any of the lines that expressed mutBP5-myc (Fig.…”

“…IGFBP-5 is the most conserved of the IGFBPs, present in all vertebrates. Like IGFBP-3, IGFBP-5 can form a complex that is retained in serum and maintains a latent reservoir of IGFs (7). IGFBP-5 is expressed early in embryonic development; its message levels are abundant in somites, indicating a potential function in myogenesis (8).…”

“…Resolved proteins were transferred to nitrocellulose membranes and probed with anti-Bax polyclonal (1:1000 dilution; BD Pharmingen), anti-Bcl-2 monoclonal (0.8 g/ml; Dako, Carpinteria, CA), or chicken anti-human IGFBP-5 (1:100; Ref. 29) antibodies overnight at 4°C. This was followed by incubation with anti-rabbit (for Bax, 1:10,000 dilution) or anti-mouse (for Bcl-2, 1:2000 dilution) IgG conjugated with horseradish peroxidase.…”

“…IGFBP-5 fragments eluted at 26 -27 min, and intact IGFBP-5 at 30 -32 min. Concentrations of IGFBP-5 in CM from IGFBP-5 expressing cells were quantitated by IGFBP-5-specific radioimmunoassay as previously described (29).…”

“…It was surprising, therefore, that the proportion of free IGF-I in serum tended to increase in Igfbp5-overexpressing mice, although this also occurred in the growth-retarded liver Igf1 and Als double-null mice (25). The proportion of different IGFBPs in serum is also important, and, in this regard, IGFBP-5 can effectively compete with IGFBP-3 for serum ternary complex formation with the acid-labile subunit in vivo (38). The demonstration of IGFBP-5-myc protein at an appropriate M r for ternary complex formation suggests that IGFBP-5 could have displaced IGFBP-3 from this complex, retaining circulating IGF in a relatively tightly bound ternary complex (39).…”
