Molecular Determinants of Pichinde Virus Infection of Guinea Pigs—a Small Animal Model System for Arenaviral Hemorrhagic Fevers

Abstract: Arenaviruses are enveloped single-strand RNA viruses that mostly have natural hosts in rodents. Upon infection of humans, several arenaviruses can cause severe hemorrhagic fever diseases, including Lassa fever that is endemic in West Africa. The virulence mechanism of these deadly arenaviruses can be studied in a safe and economical small animal model -guinea pigs infected by a non-pathogenic arenavirus Pichinde virus (PICV), a virulent strain of which can cause similar disease syndromes in guinea pigs as aren… Show more

“…The contribution of viral genomic elements to in vivo phenotype is well described for various arenaviruses (19,26). Our findings identify for the first time a critical in vivo virulence determinant in the arenavirus G2 TMD.…”

“…We have previously developed a reverse genetics system to generate infectious PICV from two plasmids that produce full-length antigenomic strands of the L and S RNAs (23). Using this reverse genetics system, we have characterized the functional mechanisms of viral virulence determinants and the biological roles of arenaviral proteins (13,(24)(25)(26)(27)(28).…”

“…Mutations in the SSP gene were created in the pCAGGS-PICV-GPC plasmid by overlapping PCR mutagenesis reactions, and the mutations were confirmed by sequencing. Plasmids used to rescue the PICV SSP mutant viruses were described previously (19). A plasmid that contains the T7 promoter-directed firefly luciferase gene was obtained from J. Nunberg (University of Montana, USA).…”
