Molecular Cytogenetic Definition of the Chicken Genome: The First Complete Avian Karyotype

Abstract: Chicken genome mapping is important for a range of scientific disciplines. The ability to distinguish chromosomes of the chicken and other birds is thus a priority. Here we describe the molecular cytogenetic characterization of each chicken chromosome using chromosome painting and mapping of individual clones by FISH. Where possible, we have assigned the chromosomes to known linkage groups. We propose, on the basis of size, that the NOR chromosome is approximately the size of chromosome 22; however, we suggest… Show more

“…Chicken (Gallus gallus, GGA) chromosome-specific DNA probes of chromosome 1-9 and Z (GGA1-9 and GGAZ) were used for comparative chromosome painting , Masabanda et al 2004. Each probe was amplified by DOP-PCR following Carter et al (1992) and hybridized to the metaphase spreads.…”

“…Karyotypes can be subdivided into large macrochromosomes including ZW-type sex chromosomes (7-10 pairs) and a large number of almost indistinguishable microchromosomes (Takagi & Sasaki 1974, Belterman & de Boer 1984 or into groups A-D (Masabanda et al 2004). …”

“…and information it provides for future research (Antin and Konieczka 2005). Together with chromosome paints, BAC clones (BPRC) have been used to define cytogenetically all chicken chromosomes (Masabanda et al 2004). Because of the nearly identical sizes of the microchromosomes in mitotic chromosome spreads, this was not previously feasible.…”

“…In this study, 10 duck loci were assigned to duck macrochromosomes, 10 loci to small macrochromosomes, and 4 loci to microchromosomes using chicken BAC clones. Much research has shown strong chromosome homology between macrochromosomes in different bird species (Masabanda et al 2004). In our research, 19 orthologous loci (CAUD023, CAUD069, AMU182, APH12, CAUD049, CAUD138, CAUD091, CAUD128, CAUD109, AMU6, CAUD111, CAUD022, APH20, CAUD038, AMU52, AMU174, CAUD044, CAUD080, and CAUD123) were assigned to the same macrochromosome or small macrochromosome in duck and chicken, respectively, 1 orthologous locus (CAUD021) was located on duck chromosome 4, but uncertain in the chicken genome.…”
