Molecular Cloning of the CA125 Ovarian Cancer Antigen

Abstract: CA125 is an ovarian cancer antigen that is the basis for a widely used serum assay for the monitoring of patients with ovarian cancer; however, detailed information on its biochemical and molecular nature is lacking. We now report the isolation of a long, but partial, cDNA that corresponds to the CA125 antigen. A rabbit polyclonal antibody produced to purified CA125 antigen was used to screen a ZAP cDNA library from OVCAR-3 cells in Escherichia coli. The longest insert from the 54 positive isolated clones had … Show more

“…Elevated serum levels of CA125 are related with cancer patients such as ovarian cancer, gastric cancer and breast cancer, etc (Bast et al, 1981). In ovarian cancer patients, CA125 could be coming down soon after operation or effect chemotherapy, recurrence of ovarian cancer may be detected through elevated levels of CA125 in the blood-long before clinical symptoms, However, elevated serum levels of CA125 are associated with benigh diseases such as Pelvic Inflammation, Endometriosis, ovarian cyst, even tuberculosis, due to the lack of a strongly specificity (O'Brien et al, 2001;Yin et al, 2001). …”

“…HE4 is a high molecular weight glycoprotein with an approximate molecular mass 80 kDa, but its secreted form is a 11 kDa protein, that explains an earlier release of biomarker in the blood stream (Schummer et al, 1999). Earlier appearance of human epidydimis secretory protein 4 in the serum in case of early stage ovarian cancer is likely to be associated with a simpler mechanism for its release (Boshell et al, 1992;Ligtenberg et al, 1992;Yin et al, 2001).…”

“…The biological functions of MSLN and MPF are not clear. MSLN binds CA125 (MUC16) that is present on many ovarian cancer cells [48,49], eluding to the suggestion that it might have a role in the metastatic spread of ovarian cancer in the peritoneal cavity [50]. Yen et al [51] recently provided new evidence that diffuse MSLN expression is associated with prolonged patient survival.…”
