Molecular Cloning, mRNA Expression and Chromosomal Localization of Mouse Angiotensin-converting Enzyme-related Carboxypeptidase (mACE2)

Abstract: We isolated two mouse cDNA clones which show significant similarities with human angiotensin-converting enzyme-related carboxypeptidase (ACE2). The cDNAs were 2746 and 1995 bp in length and seemed to arise from the same gene by alternative splicing. The longer cDNA encoded a 798-amino acid protein containing the sequence motif conserved among zinc metallopeptidases. Mouse ACE2 showed 83% identity with human ACE2. Northern blot analysis revealed that 2.8- and 2.0-kb transcripts were expressed mainly in the kidn… Show more

“…Northern blotting showed that ACE2 mRNA is expressed abundantly in endothelial cells of the heart, kidneys, and testis (37). In addition, RT-PCR and immunocytochemistry showed that ACE2 mRNA and protein are expressed on oral, nasal, and intestinal mucosa, alveolar epithelial cells in lung, stomach, skin, lymph nodes, thymus, bone marrow, spleen liver, kidney, and brain, and arterial and venous endothelial cells and arterial smooth muscle cells in many organs (38)(39)(40)(41). These sites for ACE2 expression include most of the sites for SARS-CoV replication in autopsy specimens (26).…”

“…Murine ACE2 cDNA was cloned in lentiviral vector (lenti-ACE2) essentially as described elsewhere, 18,19 and production and titration of lentiviral particles were carried out as described previously. 20,21 Lentiviral vector containing no insert was used as control, whereas a vector containing placental alkaline phosphatase reporter gene was used to characterize in vivo transduction efficiency.…”

“…The role of a recently identified receptor for SARS CoV, angiotensin-converting enzyme 2 (15), as a determinant of the host range of SARS CoV remains to be determined. The presence of angiotensin-converting enzyme 2 homologs in a wide range of species (4,12) may account for the broad host range of SARS CoV.…”
