Molecular cloning, expression and characterization of a novel feruloyl esterase enzyme from the symbionts of termite (Coptotermes formosanus) gut

Abstract: Termites play an important role in the degradation of dead plant materials and have acquired endogenous and symbiotic cellulose digestion capabilities. The feruloyl esterase enzyme (FAE) gene amplified from the metagenomic DNA of Coptotermes formosanus gut was cloned in the TA cloning vector and subcloned into a pET32a expression vector. The Ft3-7 gene has 84% sequence identity with Clostridium saccharolyticum and shows amino acid sequence identity with predicted xylanase/chitin deacetylase and endo-1,4-beta-x… Show more

“…Metagenomics [Handelsman et al, 1998] provides access to novel FAEs in natural environments and leads to the identification of enzymes with potential industrial applications [Elend et al, 2006;Voget et al, 2006]. A few novel FAEs have been identified by metagenome-based methods from guts of earthworm and termite to date [Chandrasekharaiah et al, 2011;Vleltes et al, 2010;Wheeler et al, 2010].…”

“…The ability of Tvms10a and Tvmz2a to release FA and diFA from the crude plant material was measured. However, these measurements have been carried out on phenolic acid-substituted xylan polysaccharide isolated from wheat straw and untreated crude cell wall materials (Bartolome et al, 1997;Chandrasekharaiah et al, 2011), and was found that there is a release of phenolic acids from the same substrates of wheat bran and spent grain in the presence of enzymes xylanase and feruloyl esterase (Tenkanen et al, 1991).…”

“…The purified proteins of Tvms10a and Tvmz2a were subjected to stepdialysis to obtain correct folding of recombinant FAE enzyme. The apparent molecular mass of the purified recombinant protein of Tvms10a (18.6 kDa) and Tvmz2a (31.2 kDa) was estimated using SDS-PAGE and MALDI-TOF, and resembled those of the analogous enzyme from symbiontes of termite (18.5 kDa) (Chandrasekharaiah et al, 2011) Clostridium stercorarium (33 kDa) (Donaghy et al, 2000).…”

“…N. aquilinus feeds exclusively on wood, whereas C. fulviceps consumes more nitrogen rich organic matter, including either living or decaying plant tissues 84,85 . This difference in diet may explain the metabolic pathway profiles of each termite colony.…”
