Molecular cloning, characterization and expression of a jasmonate biosynthetic pathway gene encoding allene oxide cyclase from Camptotheca acuminata

Abstract: AOC (allene oxide cyclase; EC 5.3.99.6), an essential enzyme in jasmonic acid and its methyl ester biosynthesis, was cloned from Camptotheca acuminata (named as CaAOC), a native medicinal plant species in China. CaAOC had significant similarity at the amino-acid level with AOCs from other plant species. Comparison between the sequences of the full-length cDNA and genomic DNA of CaAOC revealed that the genomic DNA of CaAOC contained an 89-bp intron and a 240-bp intron. Southern-blot analysis indicated that CaAO… Show more

“…There are indirect lines of evidence, indicating that camptothecin is involved in plant defense, and some of the biosynthetic pathway genes of camptothecin are induced by JA [17]. Previously, a full-length cDNA of AOC from C. acuminata (named CaAOC) was isolated, coding a protein of 26.8 kDa that was deduced to localize in chloroplasts [18].…”

“…The PCR amplification program was as follows: 50°C for 2 min; 95°C for 30 sec; 40 cycles of 95°C for 3 sec and 62°C for 40 sec; followed by a melting-curve program of 55°C to 85°C, with a 5-sec hold at each temperature. The gene expression patterns of all genes were normalized to an internal reference ( 18S rRNA) [53]. The relative gene expression analysis was performed using BIO-RAD IQ™5 optical system software version 2.0 with the 2 -ΔΔCt method.…”

“…Soybean GmAOC1 and GmAOC2 also exhibited abundant mRNA transcripts in roots, while GmAOC3 and GmAOC4 showed relatively higher expression in leaves and stems, respectively [7]. Camptotheca acuminata CaAOC is constitutively expressed in various organs, with the highest expression level in leaves [14]. In Arabidopsis, AtAOC1, AtAOC2, and AtAOC3 promoters exhibit high activities in the leaves, while only AtAOC3 and AtAOC4 show promoter activity in roots [15].…”
