Molecular cloning and expression of chicken cardiac troponin C.

Abstract: We have isolated a full length complementary DNA clone (pCTnC1) from a 19-day embryonic chicken heart library corresponding to cardiac troponin C (TnC). Sequence analysis demonstrated varying homologies with TnC complementary DNA clones isolated from developing chick skeletal muscle. Using pCTnC1 as a hybridization probe, we have determined that cardiac TnC is constitutively expressed in both atria and ventricles of the developing and adult heart. These data along with previous immunochemical studies of TnC ex… Show more

“…It is shown by using specific antibodies that cardiac as well as pectoral TnC exist in the pectoralis muscle of the chick embryo (Toyota and Shimada, 1981). Messenger RNA coding cardiac TnC is also shown in this embryonic muscle (Toyota et al, 1989). However, the present results about the sensitivity to CaZf, Sr"+ , and Ba2+ suggest that pectoral TnC is dominantly expressed in the cultured myotubes.…”

“…The fact that neither TnI nor Tm manifested isoform shifts during overload is consistent with previous reports. 52,53 In contrast, there are reports of reexpression of a fetal TnT isoform in human failure, 33 and we recently demonstrated a modest TnT isoform switch in hypertrophied rabbit hearts. 28 TnT exists as multiple, developmentally regulated isoforms.…”

“…Details of these clones have been described elsewhere (17)(18)(19). The cardiac nomenclature for the TnC and TnI mutants is specified according to GenBank accession numbers A27204 (20) and AAA16157 (21), respectively. Further cTnI and cTnC mutants were constructed with the Quick-Change site-directed mutagenesis kit (Stratagene) using complementary oligonucleotides containing the desired mutations into the cysteine-less background (obtained by mutating native cysteines to C35S͞C84S for cTnC and C80I͞C97Sfor cTnI).…”
