Molecular Choreography and Structure of Ca2+ Release-Activated Ca2+ (CRAC) and KCa2+ Channels and Their Relevance in Disease with Special Focus on Cancer

Abstract: Ca2+ ions play a variety of roles in the human body as well as within a single cell. Cellular Ca2+ signal transduction processes are governed by Ca2+ sensing and Ca2+ transporting proteins. In this review, we discuss the Ca2+ and the Ca2+-sensing ion channels with particular focus on the structure-function relationship of the Ca2+ release-activated Ca2+ (CRAC) ion channel, the Ca2+-activated K+ (KCa2+) ion channels, and their modulation via other cellular components. Moreover, we highlight their roles in healt… Show more

“…As shown in Figure 3 , in the model, customer experience is the prevariable of brand loyalty. Brand loyalty is divided into two dimensions: behavior and attitude loyalty, and customer consumption (CS) is introduced to expand this framework [ 11 , 12 ]. Measured by consumers' share of purchases of a particular brand relative to overall usage or spending on products in the same category.…”

“…Several reports have already provided evidence of an interplay of Ca 2+ and Ca 2+ regulated potassium (K + ) ion channels such as Ca V 2.3 with BK channels [ 9 ], Orai1 with the ether a-gogo K + channel hEag1 [ 10 ] or BK channels [ 11 , 12 ], Orai1 [ 13 ], or TRPV6 [ 14 ] with the small conductance Ca 2+ -activated activated K + channels SK4 and Orai1 with its homolog SK3 [ 13 , 15 , 16 , 17 , 18 , 19 ]. The interplay of the Ca 2+ ion channel Orai1 with the Ca 2+ -activated K + ion channel SK3 is currently best studied in breast cancer cells [ 16 , 20 , 21 ].…”

“…We further discovered that one critical determinant for this co-regulation is the CaM binding site in SK3, because Orai1 overexpression decreased the interaction of SK3 with CaM as well as the CaM MUT s. Moreover, SK3 K + currents abolished by the CaM MUT were partially restored by co-expression of Orai1. At the level of Orai1, an intact pore and both cytosolic strands, except the first 26 aa, are indispensable for the co-regulation of SK3 and Orai1 [112,301].…”
