Molecular Characterization of an Alpha Interferon Receptor 1 Subunit (IFNaR1) Domain Required for TYK2 Binding and Signal Transduction

Abstract: Binding of alpha interferon (IFN␣) to its receptors induces rapid tyrosine phosphorylation of the receptor subunits IFNaR1 and IFNaR2, the TYK2 and JAK1 tyrosine kinases, and the Stat1 and Stat2 transcription factors. Previous studies have demonstrated that TYK2 directly and specifically binds to and tyrosine phosphorylates IFNaR1 in vitro. We now report a detailed analysis of the TYK2 binding domain on the IFNaR1 subunit. First, we used an in vitro binding assay to identify the TYK2 binding motif in IFNaR1 as… Show more

“…The primary antibodies and ratios used were as follows: PSA (DAKO), 1:1,000; AR (Santa Cruz Biotechnology), 1:2,000; CDK6 (Santa Cruz Biotechnology), 1:500; hemagglutinin (HA; Covance), 1:2,000; ␤-actin (Sigma), 1:500; and insulin-like growth factor binding protein-2 (Upstate Biotechnology, Lake Placid, NY), 1:2,000. HA epitope-tagged mutants of CDK6 were constructed by using the overlapping PCR technique with appropriate primers (14). The PCR samples were thermocycled for 30 s at 95°C, at 55°C, and at 72°C.…”

“…A bacterial expression vector for GST-IFNAR1 WT protein [12,13] was a generous gift of Dr. John J. Krolewski (University of California, Irvine, CA). The pRC-Tyk2 expression vector was described elsewhere [14].…”

“…For example, Tyk2 interacts with a domain of IFN-␣R␣, the ␣-chain of the IFN-␣R (also designated as IFNAR1 or IFN-␣R1), which has only distant homology with the box 1 or box 2 motifs (25,26). Jak1 is activated by multichain receptors such as the IL-6 group of cytokines (IL-6, leukemia inhibitory factor, ciliary neurotrophic factor, oncostatin M), IFN-␣, IFN-␥, IL-10, and those cytokine receptors that belong to the IL-2 family (IL-2, IL-4, IL-7, IL-9, and IL-15) (19,20,(27)(28)(29)(30)(31)(32)(33).…”
