Molecular characterization and RNA interference responses of the lethal giant larvae gene in Diabrotica virgifera virgifera adults

Abstract: High specificity for silencing target genes and single-copy target genes that yield clear phenotypes are two important factors for the success of RNA interference (RNAi). The lethal giant larvae (Lgl) gene appears to be an ideal gene for RNAi because RNAi can effectively suppress its expression and results in molting defects and mortality in Tribolium castaneum.

“…Lethal giant larvae seems to be encoded by a single copy gene in insects such as Ostrinia nubilalis , Diabrotica virgifera virgifera , Tribolium castaneum , and Drosophila melanogaster (Ohshiro et al ., 2000; Xiao et al ., 2014; Cooper et al ., 2021). Lgl was initially identified to be a tumor suppressor in Drosophila , of which l(2)gl mutant flies died at the larval stage and showed cancer‐like defects with uncontrolled growth of larval brain neuroblasts and imaginal discs (Ohshiro et al ., 2000).…”

“…Silencing TcLgl by RNA interference (RNAi) in early larvae and late larvae resulted in 100% and 80% mortality, respectively. However, no phenotypes were observed when this gene was suppressed with RNAi in O. nubilalis , and suppression of DvLgl did not yield an observable phenotype in D. v. virgifera adults (Cooper et al ., 2021). Thus, the function of Lgl is still unclear in some insect species.…”
