Molecular characterization and phylogenetic utility of the rDNA external transcribed spacer region in Stylosanthes (Fabaceae)

Abstract: Supplementary Material The nucleotide sequence of the ribosomal external transcribed spacer (ETS) region of Stylosanthes mexicana was determined and used to evaluate its potential for examination of intra- and inter-specific relationships in Stylosanthes, as compared to the use of the internal transcribed spacer (ITS) region. The entire ETS region comprises 1,145 bp and is composed of a region of non-repetitive sequences consisting of three subregions with organizational and nucleotide-sequence conservation, a… Show more

“…Sequence-tagged sites (STS) were used to identify progenitor species for S. scabra (Liu and Musial 1997). Investigation of ribosomal DNA internal transcribed spacers (rDNA ITS) has been used to detect variation in the S. guianensis species complex and in Stylosanthes species (Vander Stappen et al 2003). Simple sequence repeats (SSR) are available for three species of Stylosanthes: S. guianensis Santos et al 2009a;Santos-Garcia et al 2012), S. capitata (Santos et al 2009b), and S. macrocephala (Santos et al 2009c).…”

“…The lower CI and RI of the ETS data (0.810 and 0.742) compared with the trnK / matK data (0.825 and 0.824) indicated a higher level of homoplasy; this was consistent with the higher rate at which the variable sites in this region evolved compared with the trnK / matK sequences (1.4 steps per site vs. 1.2 steps per site, respectively; Table 2). This pattern has also been observed in other plant genera (Baldwin & Markos, 1998; Bena et al ., 1998; Vander Stappen, Marant & Volckaert, 2003).…”

“…The psbA-trnH regions have been used at the interspecific level in the Lamiaceae (Albaladejo et al 2005;Gobert et al 2006). Similarly, the ETS region of nuclear ribosomal DNA has been found to evolve faster and consequently have greater variability than the internal transcribed spacer (ITS) region, which is more frequently used, making ETS particularly useful for studies below the generic level (Baldwin and Markos 1998;Linder et al 2000;Stappen et al 2003;Wilson et al 2012).…”
