Molecular characterization and phylogenetic relationships of two new bipartite begomovirus infecting malvaceous plants in Yucatan, Mexico

Abstract: Sida acuta and Corchorus siliquosus plants showing yellow mosaic and yellow vein symptoms, respectively, were collected in the Yucatan Peninsula, Mexico. Total DNA was isolated from both plant species and used for the amplification, cloning, and sequencing of the Begomovirus genome. Nucleotide comparison of the complete DNA-A component isolated from S. acuta and C. siliquosus confirmed the presence of two distinct begomoviruses species. Based on phenotypic symptoms observed in infected field plants, the names … Show more

“…The malvaceous plants in the New World are affected by bipartite begomoviruses [27-29] betasatellites not apparently occurring in the New World [7]. In the Old World begomoviruses infecting okra are invariably associated with betasatellites and are, for the few which have been characterized, monopartite--lacking the DNA B component.…”

“…Primers used to amplify the complete DNA-A component were prMxF-496 and prMxR-311 [6], and prAV324 and prAC889 [7]. To amplify the DNA-B component, primers prBV1855 and prBC656 were used to obtain a 1.4 kbp fragment comprising the CR and flanking sequences [8].…”

“…These results are in accordance with the pairwise sequence identity data (Table 1). No close relationship was found between CoMoV and other corchorus-infecting viruses reported from Mexico, India or Vietnam [19,20,23].…”

“…In these areas, it grows either as a ruderal or an invading weed that infests pasture preferably, but it may also occur in agricultural crops such as cotton, cassava, maize and beans [18,28]. Begomoviruses have been isolated from different species of Corchorus collected in Mexico, Vietnam and India [16,17,19,20,23]; however, Corchorus spp. have not yet been described as begomovirus hosts in Brazil.…”
