Molecular Characterization and Immunological Evaluation of Truncated Babesia microti Rhoptry Neck Protein 2 as a Vaccine Candidate

Abstract: Babesia microti is a protozoan that infects red blood cells. Babesiosis is becoming a new global threat impacting human health. Rhoptry neck proteins (RONs) are proteins located at the neck of the rhoptry and studies indicate that these proteins play an important role in the process of red blood cell invasion. In the present study, we report on the bioinformatic analysis, cloning, and recombinant gene expression of two truncated rhoptry neck proteins 2 (BmRON2), as well as their potential for incorporation in … Show more

“…The inhibition of erythrocyte invasion by rBMSA1 and rBMSA5-1-1 was evaluated using flow cytometry (MoFloXDP, Beckman, USA). The difference in the relative infection rate between the experimental and the control groups was performed as previously described [ 17 , 18 ]. The relative invasion rate=infection rate of the experimental group/infection rate of the control group.…”

“…Thereafter, 40 μl of ethidium bromide was added to the mixture and further incubated for 0.5 h at room temperature in the dark. The infection rates (the ratio of positive cells versus the total number of cells) were calculated using flow cytometry (MoFlo XDP), and 50,000 cells were collected per sample [ 17 , 18 ]. Three replicates of each concentration per sample were analysed twice to ensure data accuracy.…”
