Molecular Characterization and Expression of two Ovarian Lipoprotein Receptors in the Rainbow Trout, Oncorhynchus mykiss 1

Abstract: Partial cDNAs encoding two lipoprotein receptors were isolated and sequenced from the ovary in the rainbow trout (rt), Oncorhynchus mykiss. One of the cDNAs (rt-LPR) contained the 5 domains characteristic of receptors belonging to the low-density lipoprotein receptor superfamily. The second cDNA (rt-LP[OS]R) was similar to rt-LPR but lacked 105 base pairs encoding the O-linked sugar domain. The deduced amino acid sequences of the rt-LPR and rt-LP[OS]R had between 75% and 80% identity with very-low-density lipo… Show more

“…The molecular masses of VtgAar and Lrp13 were estimated as described above. expressed in ovary, lr8 Ϫ and lr8 + gene expression has been detected by RT-PCR in almost all somatic tissues of several other egg-laying vertebrates ( 13,19,20,44,45 ). Less sensitive methods of detection (i.e., Northern blotting) in these same studies failed to detect lr8 Ϫ gene transcripts in somatic tissues, indicating that such ectopic expression is minor.…”

“…We reported the molecular identity of a white perch Vtgr that is orthologous to mammalian VLDLR ( 18 ). This Vtgr is termed "Lr8 Ϫ ", because it is a spliced variant gene transcript of vldlr that does not encode the O -linked sugar domain, as is characteristic of this form of Vldlr in fi shes and chickens ( 19,20 ). Herein, we refer to these Vldlr forms as Lr8+ and Lr8 Ϫ , based on the presence or absence of the O -linked sugar domain, respectively.…”

“…It has been established that in oviparous vertebrates, Vtgr mediates the entrance of Vtg into oocytes, although in birds and amphibians this receptor may also play a part in the incorporation of very low density lipoproteins (VLDL) [3], and this notion has recently been adopted for fish [4,5]. With the advent of cloning technology, identification and sequencing of the vgtr has been reported for a handful of fish species, which maintain highly conserved regions; however, two cDNA isoforms have been reported in rainbow trout [3,6], blue tilapia [7], and sole [8].…”

The isoforms consist of a larger cDNA sequence containing an O-glycan domain and a shorter one devoid of this region.…”

“…In the fish ovary, lipoproteins (including Vtgs) are presumably sequestered via receptor-mediated uptake by the developing oocytes (17,45,65). Vtgs, the principal carriers of retinoids and carotenoids, are female-specific hepatically synthesized lipoproteins.…”
