SUMMARYThe melanocortin 5 receptor (MC5R) plays a key role in the regulation of exocrine secretion in mammalian species. This receptor has also been characterized in some fish species but its function is unknown. We report the molecular and pharmacological characterization, as well as the tissue expression pattern, of sea bass MC5R. Cloning of five active alleles showing different levels of sensitivity to endogenous melanocortin and one non-functional allele demonstrate the allelic complexity of the MC5R locus. The sea bass receptor was activated by all the melanocortins tested, with ACTH and desacetyl-MSH and -MSH showing the lowest efficiency. The acetylation of the MSH isoforms seems to be critical for the effectiveness of the agonist. Agouti-related protein had no effect on basal or agonist-stimulated activation of the receptor. SbMC5R was mainly expressed in the brain but lower expression levels were found in several peripheral tissues, including liver. Progressive fasting did not induce up-or downregulation of hypothalamic MC5R expression, suggesting that central MC5R is not involved in the regulation of food intake in the sea bass. MTII, a sbMC5R agonist, stimulated hepatic lipolysis in vitro, measured as free fatty acid release into the culture medium after melanocortin agonist exposure of liver fragments, suggesting that MC5R is involved in the regulation of hepatic lipid metabolism. Taken together, the data suggest that different allelic combinations may confer differential sensitivity to endogenous melanocortin in tissues where MC5R is expressed and, by extension, in hepatic lipid metabolism.

“…In C57BL/6 mice, the effect of estrogen on the gonadotrope is exerted mainly through the activation of Esr1 in the pituitary and Esr2 can partially offset the absence of Esr1 in gonadotropes in female mice by pituitary-specific Esr1 knockout (Sanchez-Criado et al, 2012). In the sea bass, in situ hybridization studies demonstrated that esr1 was profusely expressed in the main neuroendocrine areas, such as the preoptic area and hypothalamus and also was expressed in the Fshβ and Lhβ cells, suggesting involvement of Esr1 in the modulation of the hypophysiotropic neurons and direct control of gonadotropin hormone synthesis by estrogens (Muriach et al, 2008b). In the present study, the significant positive correlation between esr2a and lhβ in the brain of female G. rarus suggests that the effects of EE2 on the gonadotrope might be exerted mainly through activation esr2a rather than esr1 in mammal species and other teleost species.…”

This study was conducted to test the sensitivity to gonadal steroids of the systems regulating food intake in sea bass. Animals were treated with silastic implants containing 17-β-estradiol or testosterone. Self-feeding was recorded for 31 days using computerized demand feeders and unfed-pellet recovery systems. Both steroids strongly decreased selffeeding levels, feed efficiency and specific growth rates. The linear growth of fish treated with testosterone was higher than in 17-β-estradiol treated fish. In the second experiment, fish were treated with lower 17-β-estradiol doses and 11-keto-androstenedione, a precursor of the main fish androgen (11-keto-testosterone). The results demonstrated a dose-response effect of estrogen and no effect of non-aromatizable androgens on food intake or growth performance. The inhibitory effect of testosterone on food intake seems to be mediated by its aromatization to estradiol, while linear growth promotion is mediated by the androgen "per se". Data suggest that gonadal steroids may be involved in the seasonal feeding pattern of sea bass. The results demonstrate the sensitivity of the mechanisms regulating food intake to estrogenic compounds and point to the risk of including feed containing estrogenic substances in fish diets as well as the risk involved in exposure to "estrogenic environments".
